1. A method for extending the half-life of a mRNA transcript comprising:
- (a) providing a selected DNA sequence having a positive retroregulatory element ligated at the 3'"'"' end thereof, said positive retroregulator when transcribed into said mRNA transcript having an inverted repeat sequence capable of forming a stem and loop structure having a Δ
G°
of about -30.4 Kcal/mole and wherein said element functions in the 5'"'"' to 3'"'"' or 3'"'"' to 5'"'"' orientation to enhance expression in prokaryotic cells;
(b) providing a means for transcribing said selected DNA sequence and said positive retroregulatory element ligated thereto into said mRNA transcript wherein said means is selected from the group consisting of a prokaryotic cell and a prokaryotic cell free system; and
(c) transcribing said selected DNA sequence having said positive retroregulatory element ligated at the 3'"'"' end thereof into said mRNA transcript.

Abstract
The invention concerns a method for extending the half-life of mRNAs. The half-life extension is conferred upon the mRNA by a co-transcribed positive retroregulatory element which is ligated to the 3'"'"' end of the DNA sequence encoding the RNA. RNAs having an extended half-life conferred by a co-transcribed positive retroregulatory element are also claimed.

12. An mRNA sequence having an extended half-life conferred by a co-transcribed positive retroregulatory element, wherein said mRNA sequence is transcribed from a selected recombinant DNA sequence having a positive retroregulatory element which comprises a portion of the 3'"'"' flanking sequence of the B. thuringiensis crystal protein gene and wherein said portion comprises the sequence 5'"'"'-AAAACGGACATCACCTCCATTGAAACGGAGTGATGTCCGTTTT-3'"'"' and wherein said portion is not native to said DNA sequence ligated at the 3'"'"' end thereof in the 5'"'"' or the 3'"'"' to 5'"'"' orientation and wherein said element when transcribed is capable of forming a stem and loop structure having a Δ
