1. A method comprising(a) contacting a nucleic acid template with a primer capable of hybridizing to said template to form a primed template;

  • (b) contacting said primed template with a pool of random extension oligonucleotides having a length of N nucleotides where N is an integer from 4 to 9 wherein said extension oligonucleotides comprise random nucleotide positions containing any one of the nucleotides A. T, G or C and a predetermined nucleotide position consisting of one of the four different nucleotide residues A. T, G or C, each different nucleotide at said predetermined position containing a different nucleotide-specific label; and

    covalently linking with a ligase (1) said primer to an extension oligonucleotide and (2) two or more extension oligonucleotides when any of said primer and extension oligonucleotides are adjacently hybridized to said nucleic acid template, wherein said covalent linking produces a reaction product comprising at least one ligation product hybridized to said nucleic acid template.
