1. A device for amplifying a polynucleotide in a sample by conducting a polynucleotide amplification reaction, the device comprising:

  • a solid substrate fabricated to include at least one polynucleotide amplification reaction chamber, said chamber being permanently sealed with a cover and having one mesoscale cross-sectional dimension of width or depth between about 0.1 and 1,000 μ

    m, said reaction chamber comprising at least one wall having associated therewith a composition which diminishes inhibition by said wall of a polynucleotide amplification reaction in said chamber; and

    at least one port in fluid communication with said reaction chamber, for introducing said sample into said reaction chamber, said reaction chamber and at least one said port being of dissimilar cross-section.
