1. An improved method of detecting the presence or amount of a target nucleic acid in a sample suspected of containing said target nucleic acid, said method comprising the steps of:
- (a) providing in combination a medium suspected of containing said target nucleic acid and at least one nucleotide probe capable of binding to a region of said target nucleic acid; and
(b) determining whether said nucleotide probe has become bound to said target nucleic acid thereby indicating the presence or amount of said target nucleic acid, the improvement comprising said nucleotide probe having at least one nucleotide that has a substituent other than hydrogen at the 4'"'"'-position of the sugar moiety, said substituent being capable of facilitating the detection of said nucleotide probe.

This invention relates to use of oligonucleotides having at least one nucleotide that is substituted at the 4'"'"'-position of the sugar moiety with a substituent other than hydrogen as nucleotide probes in methods for detecting the presence or amount of a polynucleotide analyte in a sample suspected of containing the polynucleotide analyte. These oligonucleotides can also be packaged in diagnostic assay kits.

A kit for the detection of a polynucleotide analyte or target nucleic acid comprising in packaged combination at least one nucleotide probe capable of binding to a region of said analyte or target sequence, wherein said probe has at least one nucleatide that has a substituent other than hydrogen at the 4'"'"' position of the sugar moiety so that the probe can be labeled or bound to a support or can be provided with groups to render the probe labeled or bound to a support.
