1. An improved method of detecting the presence or amount of a target nucleic acid in a sample suspected of containing said target nucleic acid, said method comprising the steps of:

  • (a) providing in combination a medium suspected of containing said target nucleic acid and at least one nucleotide probe capable of binding to a region of said target nucleic acid; and

    (b) determining whether said nucleotide probe has become bound to said target nucleic acid thereby indicating the presence or amount of said target nucleic acid, the improvement comprising said nucleotide probe having at least one nucleotide that has a substituent other than hydrogen at the 4'"'"'-position of the sugar moiety, said substituent being capable of facilitating the detection of said nucleotide probe.
