Abstract

A culturing system and method particularly useful for producing cellular products such as viral pathogens of cells. It includes a mass transfer culture segment, stacked filter plates to adjust the medium composition, and a product removal and concentration segment. The mass transfer culture segment utilizes changed directional flow of the medium to maximize cell growth and production of product. The stacked filter plates allow addition of sterile fresh medium and removal of growth inhibitory substances.

Claims

1. A culturing system, comprising:

(a) a medium reservoir for containing a growth medium, and having inlet and outlet ports;

(b) a mass transfer device having first and second ends, said mass transfer device being selected from the group consisting of hollow fiber mass transfer devices and stacked plate filters;

(c) a pump including inlet and discharge ports for pumping the growth medium from the medium reservoir to the mass transfer device;

(d) a sterile barrier tangential flow membrane device connected to the mass transfer device, said sterile barrier tangential flow membrane device being selected from the group consisting of hollow fiber mass transfer devices and stacked plate filters;

(e) means for harvesting microorganisms grown in the mass transfer device, such harvesting means being selectively openably and closably coupled with the mass transfer device;

(f) medium flow control means comprising flow ports designated A, B, C, and D for connecting;

(i) in a first configuration, port A in flow communication with B, and port C in flow communication with port D; and

(ii) in a second configuration, port A in flow communication with port D, and port B in flow communication with port C; and

(g) conduits connecting the pump discharge port to port C, the pump inlet to the reservoir outlet port, the reservoir inlet port to port A, the mass transfer device first end to the port B and the mass transfer device second end to port D.

2. A culturing system according to claim 1, for culturing cells producing infected viruses, wherein the harvesting means comprises a vessel connected to source of chemical capable of inactivating the viruses to render them unifective, and arranged so that the chemical is selectively introduceable to the vessel.

3. A culturing system according to claim 1, wherein the sterile barrier tangential flow membrane device comprises a stacked plate filter system.

4. A culturing system according to claim 3, wherein the stacked plate filter system comprises:

(a) a support including a circumscribing frame with an array of spaced-apart and substantially parallelly aligned ribs extending between and joined at their opposite ends to the frame, so that the ribs and frame form a series of corresponding substantially parallel filtrate flow channels, and openings in the frame in liquid flow communication with the filtrate flow channels for egress of filtrate from the filtrate flow channels through the frame openings;

(b) a first filter sheet secured to a first face of the frame; and

(c) a second filter sheet secured to a second face of the frame;

wherein the first and second filter sheets together with the frame define an enclosed interior volume comprising the filtrate flow channels separated by the ribs, whereby filtrate entering the enclosed interior volume through the first and second filter sheets flows in the filtrate flow channels and is discharged from the filter element through the frame openings in liquid flow communication with the filtrate flow channels.

5. A culturing system according to claim 3, wherein the stacked plate filter system comprises:

(a) a first generally planar and rectangular filter plate having a substantially flat bottom surface, and a top surface with an upwardly extending wall circumscribingly bounding a flow channel of generally rectangular shape with a liquid inlet port at a medial part of a first side of the flow channel and a liquid outlet port at a medial part of a second side of the flow channel opposite the first side thereof, the liquid inlet port being joined in liquid flow communication with a liquid feed trough extending transversely across the first side of the flow channel, and the liquid outlet port being joined in liquid flow communication with a liquid collection trough extending transversely across the second side of the flow channel, with a plurality of spaced-apart partitions extending upwardly from the floor of the flow channel between the liquid feed trough and the liquid collection trough, the partitions being of lesser height than the wall circumscribing said flow channel and substantially parallel to each other to define a series of sub-channels extending longitudinally between the liquid feed trough and the liquid collection trough, the liquid feed trough being of progressively decreasing depth from its medial portion, in communication with the liquid inlet port, to its marginal extremities, and the liquid collection trough being of progressively decreasing depth from its medial portion, in communication with the liquid outlet port, to its marginal extremities;

(b) a foraminous support of generally rectangular shape supportively reposable at a first face thereof on the partitions of the first filter plate; and

(c) a second filter plate structurally identical to said first plate member, positioned in inverted relationship to said first plate member such that said circumscribingly bounding walls of said first and second filter plates are in abutting contact with one another, with said foraminous support between said first and second filter plates and supported by the respective partitions thereof; and

(d) filter sheets on either side of the foraminous support, interposed between the foraminous support and the supporting partitions of the respective first and second filter plates.

A culturing system according to claim 3, wherein said stacked filter plate system comprises a filter plate having a generally planar and rectangular shape with a substantially flat bottom surface, and a top surface with an upwardly extending wall circumscribingly bounding a flow channel of generally rectangular shape, with a liquid inlet port at a medial part of a first side of the flow channel and a liquid outlet port at a medial part of a second side of the flow channel opposite the first side thereof, the liquid inlet port being joined in liquid flow communication with a liquid feed trough extending transversely across the first side of flow channel, and the liquid outlet port being joined in liquid flow communication with a liquid collection trough extending transversely across the second side of the flow channel, with a plurality of spaced-apart partitions extending upwardly from the floor of the flow channel between the liquid feed trough and liquid collection trough, the partitions being of lesser height than the wall circumscribing the flow channel and substantially parallel to one another to define a series of sub-channels extending longitudinally between the liquid feed trough and the liquid collection trough, the liquid feed trough being of progressively decreasing depth from its medial portion, in communication with the liquid inlet port, to its marginal extremitites, and the liquid collection trough being of progressively decreasing depth from its medial portion, in communication with the liquid outlet port, to its marginal extremities.

7. A culturing system according to claim 1, wherein the medium flow control means comprises a switchable flow control means for controlling the direction of flow of medium through the mass transfer device.

8. A culturing system according to claim 1, wherein the medium is capable of growing host cells for viral pathogens and the harvesting means comprises a virus removal, concentration and lysis system.

9. A culturing system according to claim 1, further comprising a culturing system control means for automatically controlling medium flow.

10. A culturing system according to claim 1, further comprising means for exchanging nutrient into and out of the medium.

11. A method of culturing microorganisms in a closed culturing system to obtain a desired product, comprising the steps of:

(a) providing a mass transfer culturing system comprising;

(i) a reservoir having inlet and outlet ports;

(ii) a pump having inlet and discharge ports;

(iii) a mass transfer device with first and second ends, said mass transfer device being selected from the group consisting of hollow fiber mass transfer devices and stacked plate filters; and

(iv) flow control means comprising flow ports designated A, B, C, and D, for connecting;

(A) in a first configuration, port A in flow communication with port B, and port C in flow communication with port D; and

(B) in a second configuration, port A in flow communication with port D, and port B in flow communication with port C; and

(v) conduits connecting the pump discharge port to port C, the pump inlet to the reservoir outlet port, the reservoir inlet port to port A, the mass transfer device first end to port B, and the mass transfer device second end to port D;

(b) providing in the mass transfer device an inoculum of cells capable of producing a desired product;

(c) flowing a medium from the reservoir through the mass transfer device to expose the cells thereto;

(d) periodically switching the flow control means between the respective first and second configurations to periodically reverse the flow of medium through the mass transfer device;

(e) adjusting the composition of the medium to obtain production of the desired product, by means of a sterile barrier tangential flow membrane device selected from the group consisting of hollow fiber mass transfer devices and stacked plate filters; and

(f) filtratingly concentrating the desired product produced by the cells in the closed culturing system.

12. A method according to claim 11, comprising removing low molecular weight inhibitory substances by membrane dialysis of the medium flowed through the mass transfer device.

13. A method according to claim 11, wherein the desired product comprises a cellular pathogen.

14. A method according to claim 13, wherein the cellular pathogen comprises a virus.

15. A method according to claim 14, further comprising treating the virus to destroy the pathogenicity thereof after it has been filtratingly concentrated.

16. A method according to claim 11, wherein the desired product comprises HIV virus.

17. A method according to claim 11, wherein the desired product is a virus.

18. A culturing system comprising:

(a) a tangential flow growth device for growing a cell culture, said tangential flow growth device being selected from the group consisting of hollow fiber mass transfer devices and stacked plate filters;

(b) a flow circuit coupled to the tangential flow growth device for selectively reversibly flowing a cell growth medium therethrough in one of selected opposite flow directions, said flow circuit comprising;

(i) a cell growth medium reservoir;

(ii) a pump;

(iii) a four-way valve; and

(iv) a sterile barrier tangential flow membrane device selected from the group consisting of hollow fiber mass transfer devices and stacked plate filters, wherein the flow circuit is constructed and arranged to flow cell growth medium through the tangential flow growth device in one of two selected opposite flow directions;

(c) an auxiliary flow circuit comprising an auxiliary reversible pump, joined to the tangential flow growth device and arranged for flowing cell culture from the tangential flow growth device through the auxiliary circuit for recirculation thereof; and

(d) means for concentrating the cells in cell culture which is withdrawn from the tangential flow growth device, comprising an ultrafiltration means.

19. A culturing system, comprising:

(a) a tangential flow growth device for culturing of a cell culture;

(b) a flow circuit including a growth medium reservoir and pump for flowing growth medium through the tangential flow growth device; and

(c) a sterile barrier port means coupled to the tangential flow device for sterilely introducing or removing selected material to or from the tangential flow growth device, comprising two valves in series, each of which is independently selectively openable or closable, said valves being contiguous and in closed flow communication with one another, without any flow circuit branches therebetween.
