1. A device for separating a desired analyte from a fluid sample, the device comprising a cartridge having:
- a) a sample flow path;
b) a lysing chamber in the sample flow path, wherein the lysing chamber contains at least one filter for capturing cells or viruses from the sample as the sample flows through the lysing chamber;
c) beads disposed in the lysing chamber for rupturing the cells or viruses to release the analyte therefrom;
d) a waste chamber in fluid communication with the lysing chamber via the sample flow path for receiving the remaining sample after the sample flows through the lysing chamber;
e) an analyte flow path extending from the lysing chamber, wherein the analyte flow path diverges from the sample flow path; and
f) at least one flow controller for directing the sample into the waste chamber after the sample flows through the lysing chamber and for directing the analyte separated from the sample into the analyte flow path.

Abstract
A cartridge for separating a desired analyte from a fluid sample has a sample flow path and a lysing chamber in the sample flow path. The lysing chamber contains at least one filter for capturing cells or viruses from the sample as the sample flows through the lysing chamber. Beads are also disposed in the lysing chamber for rupturing the cells or viruses to release the analyte therefrom. An analyte flow path extends from the lysing chamber and diverges from the sample flow path. The analyte flow path preferably leads to a reaction chamber for chemically reacting and optically detecting the analyte. The cartridge also includes at least one flow controller (e.g., valves) for directing the sample into the waste chamber after the sample flows through the lysing chamber and for directing the analyte separated from the sample into the analyte flow path.

b) a first wall having a first spillover rim disposed between the waste chamber and the first sensor chamber for preventing liquid from flowing from the waste chamber into the sensor chamber until the liquid in the waste chamber reaches the level of the spillover rim.

11. The device of claim 10, wherein the cartridge further includes:

a) a second sensor chamber in fluid communication with the waste chamber; and
b) a second wall having a second spillover rim disposed between the waste chamber and the second sensor chamber, the second spillover rim being positioned at a different level relative to the bottom of the waste chamber than the first spillover rim.

12. The device of claim 1, further comprising a reaction chamber for receiving the analyte separated from the sample, wherein the reaction chamber is in fluid communication with the lysing chamber via the analyte flow path.

13. The device of claim 1, wherein the cartridge includes at least one wall defining the lysing chamber, and wherein the device further comprises a transducer for contacting an external surface of the wall to generate pressure waves in the lysing chamber.

14. (amended) The device of claim 13, wherein the transducer comprises an ultrasonic horn.

15. The device of claim 1, wherein the cartridge further includes a mixing chamber for mixing the analyte with a reagent, wherein the mixing chamber is in fluid communication with the lysing chamber via the analyte flow path.

16. The device of claim 15, wherein the cartridge further includes a reaction chamber, in fluid communication with the mixing chamber, for amplifying the analyte.

The device of claim 1, wherein the cartridge further includes a reaction chamber, in fluid communication with the lysing chamber via the analyte flow path, for amplifying the analyte and holding the analyte for optical detection, wherein the cartridge is in combination with an instrument having a heater for heating the reaction chamber and having at least one optical detector for detecting the analyte.

18. The device of claim 1, wherein the cartridge further includes:

i) a reaction chamber, in fluid communication with the lysing chamber via the analyte flow path, for amplifying the analyte;
ii) a capillary electrophoresis area in communication with the reaction chamber.

19. The device of claim 1, wherein the cartridge includes at least one wall defining the lysing chamber, the wall having a surface external to the lysing chamber for contacting a transducer to sonicate the lysing chamber.

20. The device of claim 19, wherein the wall comprises a sheet or film of polymeric material.

21. The device of claim 20, wherein the wall has a thickness in the range 0.025 to 0.1 mm.

22. The device of claim 19, wherein the wall is dome-shaped and convex with respect to the transducer.

23. The device of claim 19, wherein the wall has stiffening ribs.
