1. A method of activating expression of a developmentally silenced endogenous cellular gene in a cell;

  • the method comprising the steps of;

    (a) administering a nucleic acid to the cell, wherein the nucleic acid comprises a sequence encoding a first zinc finger protein operably linked to a promoter, wherein the first zinc finger protein is engineered; and

    (b) contacting a first target site in the endogenous cellular gene with the first zinc finger protein, wherein the Kd of the zinc finger protein is less than about 25 nM;

    wherein said contacting results in activation of expression of the endogenous cellular gene to at least about 150%.
