Abstract

This invention provides methods of retentate chromatography for resolving analytes in a sample. The methods involve adsorbing the analytes to a substrate under a plurality of different selectivity conditions, and detecting the analytes retained on the substrate by desorption spectrometry. The methods are useful in biology and medicine, including clinical diagnostics and drug discovery.

Claims

1. A probe used in laser desorption/ionization mass spectrometry comprising:

a) a metal substrate in the shape of a plate or a strip having a flat surface, wherein the flat surface of the substrate is coated with silicon oxide or titanium oxide;

b) a plurality of addressable locations on the flat surface of the substrate, wherein each addressable location comprises an adsorbent covalently attached to the coated surface,

wherein the probe is inserted into a laser desorption/ionization mass spectrometer.

2. The probe of claim 1 wherein the adsorbent is an anion exchange adsorbent.

3. The probe of claim 1 wherein the adsorbent is a cation exchange adsorbent.

4. The probe of claim 1 wherein the adsorbent is a hydrophilic adsorbent.

5. The probe of claim 1 wherein the adsorbent is a hydrophobic adsorbent.

6. The probe of claim 1 wherein the adsorbent is a metal chelate adsorbent.

7. The probe of claim 1 wherein the adsorbent is a reversible covalent interaction adsorbent.

8. The probe of claim 1 wherein the adsorbent is a biospecific adsorbent.

9. The probe of claim 1 wherein the adsorbent is silicon oxide.

10. The probe of claim 1 wherein different adsorbents are attached to different addressable locations.

11. The probe of any one of claims 1 or 2–

8 wherein the coated surface is derivatized with a bifunctional linker and wherein the bifunctional linker is further derivatized with a group that functions as the adsorbent.

12. The probe of claim 11 wherein the bifunctional linker is attached to the coated surface through an inorganic oxide functional group of the bifunctional linker.

13. The probe of claim 11 wherein the bifunctional linker is a residue of aminopropyl triethoxysilane.

14. The probe of claim 11 wherein the bifunctional linker is a residue of carbodiimide or N-hydroxysuccinimide.

15. The probe of any one of claims 1 or 2–

8 wherein a cross-linked polymer is bound to the coated surface through a functional group on the cross-linked polymer and wherein the adsorbent is bound to the cross-linked polymer.

16. The probe of claim 15 wherein the cross-linked polymer comprises cellulose, dextran, carboxymethyl dextran, polyacrylamide, or mixtures thereof.
