1. An in vitro method of making a small molecule, the method comprising the steps of:

  • (a) providing (i) a small molecule scaffold comprising a plurality of sites for functionalization covalently attached to a first oligonucleotide, and (ii) a building block provided by a reactive unit covalently attached to a second oligonucleotide capable of hybridizing to the first oligonucleotide; and

    (b) annealing the first and second oligonucleotides to each other to bring the building block and the small molecule scaffold into reactive proximity so that the building block, without the assistance of a ribosome, becomes covalently attached to the scaffold via a first one of the sites for functionalization to produce a reaction product,wherein, after step (b), the reaction product is not a nucleic acid and the first oligonucleotide comprises an oligonucleotide sequence that identifies the building block covalently attached to the scaffold during step (b).
