Abstract

Disclosed herein is a composition suitable for use in spinal surgery. The composition generally includes a reaction product or settable mixture of first and second components, the first component including a spray-dried inorganic filler, a radical donor, and a pre-polymerized vinyl polymer, and the second component including a radical scavenger, a diluent, a polymerization accelerator, and a reactive monomer. Furthermore, disclosed herein is a method of making the composition, which generally includes mixing the first and second components under conditions suitable to form a curable reaction product.

Claims

1. A composition comprising a reaction product of:

a) a first component comprising a spray-dried hydroxyapatite, a radical donor and poly(methyl methacrylate) (PMMA), wherein the spray-dried hydroxyapatite is present at about 30 wt. % to about 99.5 wt. %, based on the total weight of the first component and has a particle size of about 20 micrometers to about 35 micrometers; and

,

b) a second component comprising a radical scavenger, a diluent, a polymerization accelerator, and a reactive monomer of methyl methacrylate (MMA), and wherein the first component and second component are present in a weight ratio of about 2.2;

The composition of claim 1, wherein the first component further comprises an optional ingredient selected from the group consisting of an antibiotic, a cytostatic agent, an analgesic agent, a disinfectant, a preservative, a growth factor, a proliferative factor, a protein, a peptide, a biopolymer, a dye, an imageable marker, and mixtures thereof.

3. The composition of claim 2, wherein the imageable marker is present in the first component and comprises up to about 30 wt. % of the first component, based on the total weight of the first component.

4. The composition of claim 3, wherein the imageable marker is present in the first component at up to about 10 wt. % of the first component, based on the total weight of the first component.

5. The composition of claim 1, wherein the second component further comprises an optional ingredient selected from the group consisting of a dye, an admixture of proteins, a chemotherapeutic, a drug, an antibiotic, and mixtures thereof.

6. The composition of claim 1, wherein the poly(methyl metacrylate) comprises beads having an average particle size of about 20 micrometers (μ

m) to about 55 μ

m.

7. The composition of claim 1, wherein the diluent is selected from the group consisting of polyethylene glycol (PEG), an ester of mellitic acid, and mixtures thereof.

8. The composition of claim 7, wherein the diluent has a molecular weight such that the diluent is a liquid at standard temperature and pressure.

9. The composition of claim 7, wherein the PEG has a molecular weight of about 100 to about 1000 Daltons.

10. The composition of claim 1, wherein the first component comprises about 35 wt. % to about 99.5 wt. % of the hydroxyapatite, based on the total weight of the first component.

11. The composition of claim 1, wherein the first component comprises about 0.5 wt. % to about 5 wt. % of the radical donor, based on the total weight of the spray-dried hydroxyapatite and the poly(methyl methacrylate) (PMMA).

12. The composition of claim 11, wherein the first component comprises about 0.6 wt. % to about 3 wt. % of the radical donor, based on the total weight of the hydroxyapatite and the poly(methyl methacrylate) (PMMA).

13. The composition of claim 12, wherein the first component comprises about 0.9 wt. % to about 2 wt. % of the radical donor, based on the total weight of the spray-dried hydroxyapatite and the poly(methyl methacrylate) (PMMA).

14. The composition of claim 1, wherein the first component comprises about 0.5 wt. % to about 99.5 wt. % of poly(methyl methacrylate) (PMMA), based on the total weight of the first component.

15. The composition of claim 14, wherein the first component comprises about 50 wt. % to about 90 wt. % of poly(methyl methacrylate) (PMMA), based on the total weight of the first component.

16. The composition of claim 14, wherein the first component comprises about 30 wt. % to about 70 wt. % of poly(methyl methacrylate) (PMMA), based on the total weight of the first component.

17. The composition of claim 1, wherein the radical scavenger comprises about 5 parts per million parts of the second component (ppm) to about 500 ppm.

18. The composition of claim 17, wherein the radical scavenger comprises about 30 parts per million parts of the second component (ppm) to about 400 ppm.

19. The composition of claim 1, wherein the second component comprises about 1 wt. % to about 90 wt. % of the diluent, based on the total weight of the second component.

20. The composition of claim 1, wherein second component comprises about 0.1 wt. % to about 3 wt. % of the polymerization accelerator based on the total weight of the second component.

21. The composition of claim 20, wherein second component comprises about 0.2 wt. % to about 2 wt. % of the polymerization accelerator, based on the total weight of the second component.

22. The composition of claim 21, wherein second component comprises about 0.3 wt. % to about 0.4 wt. % of the polymerization accelerator, based on the total weight of the second component.

23. The composition of claim 1, wherein, the first component and second component are present in a weight ratio of about 2.5:

24. The composition of claim 1, wherein the first component comprises about 30 wt. % to about 50 wt. % of the hydroxyapatite, based on the total weight of the first component.

25. The composition of claim 1, wherein the radical donor is benzoyl peroxide (BPO).

26. The composition of claim 1, wherein the radical scavenger is hydroquinone monomethylether.

27. The composition of claim 1, wherein the polymerization accelerator is dimethylparatoluidine (DMPT).

28. The composition of claim 1, wherein the radical donor is benzoyl peroxide (BPO), the radical scavenger is hydroquinone monomethylether, and the polymerization accelerator is dimethylparatoluidine (DMPT).

29. The composition of claim 1, wherein the second component comprises about 10 wt. % to about 99 wt. % of the reactive monomer methyl methacrylate (MMA), based on the total weight of the second component.

30. The composition of claim 29, wherein the second component comprises about 40 wt. % to about 95 wt. % of the reactive monomer, based on the total weight of the second component.

31. The composition of claim 30, wherein the second component comprises about 60 wt. % to about 90 wt. % of the reactive monomer, based on the total weight of the second component.

32. A composition comprising a reaction product of:

a) a first component comprising a spray-dried hydroxyapatite, benzoyl peroxide (BPO), barium sulfate (BaSO4), and poly(methyl methacrylate) (PMMA)), wherein the spray-dried hydroxyapatite is present at about 30 wt. % to about 99.5 wt. %, based on the total weight of the first component and has a particle size of about 20 micrometers to about 35 micrometers, and

b) a second component reactive with the first component, the second component comprising hydroquinone monomethylether, a diluent, dimethylparatoluidine (DMPT), and methyl methacrylate (MMA),

wherein the first component and second component are present in a weight ratio of about 2.2;

33. A composition comprising a reaction product of:

a) a first component comprising;

about 30 wt. % to about 50 wt. % of a spray-dried hydroxyapatite, based on the total weight of the first component, wherein the spray-dried hydroxyapatite has a particle size of about 20 micrometers to about 35 micrometers;

about 30 wt. % to about 70 wt. % of poly(methyl methacrylate) (PMMA), based on the total weight of the first component,

about 0.6 wt. % to about 3 wt. % of a radical donor, based on the total weight of the spray-dried hydroxyapatite and the poly(methyl methacrylate) (PMMA),

up to about 30 wt. % of an imageable marker, based on the total weight of the first component; and

,

b) a second component comprising;

about 10 wt. % to about 99 wt. % of a reactive monomer of methyl methacrylate (MMA), based on the total weight of the second component,

about 0.3 wt. % to about 0.4 wt. % of a polymerization accelerator, based on the total weight of the second component, and,

about 30 parts per million parts of the second component (ppm) to about 400 ppm of a radical scavenger, wherein the first component and second component are present in a weight ratio of about 2.2;

34. A composition comprising a reaction product of:

a) a first component comprising a spray-dried hydroxyapatite, benzoyl peroxide (BPO), barium sulfate (BaSO4), and poly(methyl methacrylate) (PMMA), wherein the spray-dried hydroxyapatite is present at about 30 wt. % to about 99.5 wt. % and has a particle size of about 20 micrometers to about 35 micrometers, based on the total weight of the first component and wherein barium sulfate (BaSO4) is present in the first component at up to about 10 wt. % of the first component, based on the total weight of the first component, and,

b) a second component comprising hydroquinone monomethylether, dimethylparatoluidine (DMPT), and methyl methacrylate (MMA).

35. A composition comprising a reaction product of:

a) a first component comprising a spray-dried hydroxyapatite, benzoyl peroxide (BPO), and poly(methyl methacrylate) (PMMA), wherein the spray-dried hydroxyapatite is present at about 30 wt. % to about 99.5 wt. %, based on the total weight of the first component and has a particle size of about 20 micrometers to about 35 micrometers and wherein the poly(methyl methacrylate) comprises beads having an average particle size of about 20 micrometers to about 55 micrometers, and

b) a second component comprising hydroquinone monomethylether, dimethylparatoluidine (DMPT), and methyl methacrylate (MMA).
