Abstract

The present invention relates to a three-dimensional system, and compositions obtained therefrom, wherein individual layers of the system comprise channels divided longitudinally into two compartments by a centrally positioned membrane, and wherein each compartment can comprise a different cell type.

Claims

1. A three-dimensional system comprising at least two layers, wherein each layer comprises channels divided longitudinally into two compartments by a centrally positioned membrane, wherein each compartment includes a different cell type, and wherein the membrane has pores sized to enable fluid communication between the compartments while substantially preventing the passage of cells.

2. The system of claim 1, wherein one compartment of at least one channel comprises parenchymal cells

3. The system of claim 1, wherein one compartment of at least one channel comprises vasculature.

4. The system of claim 1, wherein one compartment of at least one channel comprises parenchymal cells and one compartment of at least one channel comprises vasculature.

5. The system of claim 4, wherein the parenchymal cells and the vasculature are present in separate compartments of the same channel.

The system of claim 4, wherein the parenchymal cells are selected from the group consisting of smooth or skeletal muscle cells, myocytes, fibroblasts, chondrocytes, adipocytes, fibromyocytes, ectodermal cells, ductile cells, skin cells, hepatocytes, kidney cells, pancreatic islet cells, intestinal cells, nerve cells, and osteoblasts.

7. The system of claim 1, wherein the cells in the compartments align vertically with cells of the same type in an adjacent layer.

8. The system of claim 1, further comprising a support element for each layer, wherein the support element of at least one layer is made of a nondegradable material.

9. The system of claim 8, wherein the nondegradable material is a polymer.

10. The system of claim 9, wherein the polymer is PolyDiMethylSiloxane or PolyMethylMethacrylate.

11. The system of claim 1, further comprising a support element for each layer, wherein the support element of at least one layer is made of a biodegradable material.

12. The system of claim 11, wherein the biodegradable material is PolyCaproLactone.

13. The system of claim 11, wherein the biodegradable material is a biorubber.

14. The system of claim 1, wherein one or more layers further comprise a sealant.

15. The system of claim 1, wherein the membrane is a semi-permeable membrane.

16. The system of claim 1, wherein the membrane is made of a biologically compatible nondegradable material.

17. The system of claim 16, wherein the biologically compatible nondegradable material is selected from the group consisting of PolyDiMethylSiloxane, PolyMethylMethacrylate, PolyEtherSulfone, and PolyCarbonate.

18. The system of claim 1, wherein the membrane is made of a biologically compatible degradable material

19. The system of claim 18, wherein the biologically compatible degradable material is Polylactide-Co-Glycolide or PolyCaproLactone.

20. The system of claim 18, wherein the biologically compatible degradable material is a biorubber.

21. The system of claim 1, wherein the membrane pores have cross-sections ranging from about 100 nanometers to about 2 microns.

22. The system of claim 21, wherein the membrane further comprises a solid material selected from the group consisting of collagen, fibronectin, laminin and self-assembling peptides.

23. The system of claim 22, wherein the solid material is contained within at least one pore of the membrane.

24. The system of claim 1, wherein at least one layer further comprises connections for inflow and outflow.

25. The system of claim 1, further comprising a bridge connecting two sections of the same compartment.

26. The system of claim 1, further comprising a bridge connecting a compartment of one channel to a compartment of another channel.

27. The system of claim 26, wherein the bridge connects compartments having same cell types.

28. The system of claim 26, wherein the bridge connects compartments having primary cells in one of the connected compartments and compatible cells in the other connected compartment for providing support to the primary cells.

29. The system of claim 28, wherein the primary cells are parenchymal cells.

30. The system of claim 29, wherein the parenchymal cells are selected from the group consisting of smooth or skeletal muscle cells, myocytes, fibroblasts, chondrocytes, adipocytes, fibromyocytes, ectodermal cells, ductile cells, skin cells, hepatocytes, kidney cells, pancreatic islet cells, intestinal cells, nerve cells, and osteoblasts.

31. The system of claim 28, wherein the compatible cell type is a stromal or connective cell.

32. The system of claim 28, wherein the compatible cell type is a fibroblast.

33. The system of claim 28, wherein the compatible cell types consist of hepatocytes and fibroblasts.

34. A device comprising a three-dimensional system of at least two layers, wherein each layer comprises channels divided longitudinally into two compartments by a centrally positioned membrane, wherein each compartment includes a different cell type, and wherein the membrane has pores sized to enable fluid communication between the compartments while substantially preventing the passage of cells;

and a pumping means for circulating fluid through the device.

35. The device of claim 34, wherein the device is extracorporeal.

36. The device of claim 34, wherein the device is implanted into a subject.

37. A three-dimensional system comprising at least two layers, wherein each layer comprises channels divided longitudinally into two compartments by a centrally positioned membrane, wherein each compartment comprises a different cell type, and wherein at least one channel comprises parenchymal cells in one compartment and vasculature in the other compartment.
