Abstract

This invention pertains to methods and compositions for the diagnosis and treatment of cardiovascular conditions. More specifically, the invention relates to isolated molecules that can be used to diagnose and/or treat cardiovascular conditions including cardiac hypertrophy, myocardial infarction, stroke, arteriosclerosis, and heart failure.

Claims

1. A method of diagnosing a cardiovascular condition in a subject, said method comprising:

a) determining a level of secreted Fit-1 polypeptide in a biological sample comprising serum from a subject; and

b) determining if the level of secreted Fit-1 polypeptide in the sample is increased relative to a predetermined value,

wherein the presence of a level that is increased relative to the predetermined value is diagnostic of the condition in the subject.

2. The method of claim 1, wherein the condition is a cardiovascular condition selected from the group consisting of myocardial infarction, stroke, arteriosclerosis, and heart failure.

3. The method of claim 1, wherein the condition is cardiac hypertrophy.

6. The method of claim 1, wherein the sample comprises blood from the subject.

7. The method of claim 1, wherein the step of determining comprises contacting the sample with an antibody that selectively binds the Fit-1 polypeptide.

12. The method of claim 1, wherein the subject is a human.

14. The method of claim 7, wherein the antibody is a monoclonal antibody that binds the human Fit-1 polypeptide.

16. The method of claim 1, further comprising administering a treatment for the condition to the subject.

17. The method of claim 16, wherein the treatment is administration of a HMG-CoA reductase inhibitor.

4. A method for monitoring a cardiovascular condition in a subject over time, wherein said cardiovascular condition is characterized by the presence of increased serum levels of soluble Fit-1 polypeptide, said method comprising determining levels of secreted Fit-1 polypeptide in a plurality of biological samples comprising serum obtained from the subject over time.

5. The method of claim 4, wherein the step of determining comprises contacting the samples with an antibody that selectively binds the Fit-1 polypeptide.

8. The method of claim 4, wherein the levels of secreted Fit-1 polypeptide over time indicate whether the condition is regressing, progressing, or at onset.

9. The method of claim 4, wherein the condition is a cardiovascular condition selected from the group consisting of myocardial infarction, stroke, arteriosclerosis, and heart failure.

10. The method of claim 4, wherein the condition is cardiac hypertrophy.

11. The method of claim 4, wherein each of the samples comprises blood from the subject.

13. The method of claim 4, wherein the subject is a human.

15. The method of claim 5, wherein the antibody is a monoclonal antibody that binds the human fit-1 polypeptide.

18. The method of claim 5, further comprising administering a treatment for the condition to the subject.

19. The method of claim 18, wherein the treatment is administration of a HMG-CoA reductase inhibitor.

20. A method for monitoring a cardiovascular condition selected from the group consisting of myocardial infarction, stroke, arteriosclerosis, and heart failure in a subject over time, said method comprising determining levels of secreted Fit-1 polypeptide in a plurality of biological samples comprising serum obtained from the subject over time.

21. The method of claim 20, wherein the step of determining comprises contacting the samples with an antibody that selectively binds the Fit-1 polypeptide.

22. The method of claim 21, wherein the antibody is a monoclonal antibody that binds the human Fit-1 polypeptide.

23. The method of claim 20, wherein the levels of secreted Fit-1 polypeptide over time indicate whether the condition is regressing, progressing, or at onset.

24. The method of claim 20, wherein each of the samples comprises blood from the subject.

25. The method of claim 20, wherein the subject is a human.

26. The method of claim 20, wherein the condition is cardiac hypertrophy.

27. The method of claim 20, wherein the condition is heart failure.

28. The method of claim 20, further comprising administering a treatment for the condition to the subject.

29. The method of claim 28, wherein the treatment is administration of a HMG-CoA reductase inhibitor.
