Abstract

The invention includes methods and compositions for remodeling a peptide molecule, including the addition or deletion of one or more glycosyl groups to a peptide, and/or the addition of a modifying group to a peptide.

Claims

1. A covalent conjugate formed between a poly(ethylene glycol) and a Factor VIII peptide, said peptide having the formula:

2. The covalent conjugate of claim 1, wherein said Factor VIII peptide is a Factor VIII variant wherein the Factor VIII variant further comprises a B-domain deletion.

3. The covalent conjugate of claim 1, wherein said Factor VIII variant further comprises amino acids 1-740 linked to amino acids 1649 to 2332 by at least 3 amino acid residues.

4. The covalent conjugate of claim 1, said method further comprising:

(c) prior to step (b), removing a group added to said saccharide during post-translational modification.

5. The covalent conjugate of claim 4, wherein said group is a member selected from phosphate, sulfate, carboxylate and esters thereof.

6. The covalent conjugate of claim 1, wherein said peptide has the formula:

7. The covalent conjugate of claim 1, wherein said poly(ethylene glycol) is a monomethoxy-poly(ethylene glycol).

8. The covalent conjugate of claim 1, wherein said poly(ethylene glycol) is a member selected from linear poly(ethylene glycol) and branched poly(ethylene glycol).

9. The covalent conjugate of claim 1, wherein said poly(ethylene glycol) has a molecular weight distribution that is essentially homodisperse.

10. The covalent conjugate of claim 1, wherein said at least one glycosyltransferase is selected from the group consisting of ST3Gal1, ST3Gal3, ST6GalNAcI CST-II and combinations thereof.

11. The covalent conjugate of claim 1, said method further comprising:

(d) contacting said covalent conjugate with a sialic acid donor and a sialyltransferase under conditions suitable for said sialyltransferase to transfer a sialic acid residue onto said covalent conjugate, thereby transferring a sialic acid moiety onto said covalent conjugate.

12. The covalent conjugate of claim 1, wherein said peptide has the formula:

q is selected from the integers between 1 and 20, and when q is three or greater (mannose)_q is selected from linear and branched structures.

and i and k are independently selected from the integers 0 and 1, with the proviso that at least one of i and k is 1, and when k is 1, g, h, and j are 0.

19. A covalent conjugate formed between a poly(ethylene glycol) and a Factor VIII peptide, said peptide having the formula:

20. The covalent conjugate of claim 19, wherein said Factor VIII peptide is a Factor VIII variant wherein the Factor VIII variant further comprises a B-domain deletion.

21. The covalent conjugate of claim 20, wherein said Factor VIII variant further comprises amino acids 1-740 linked to amino acids 1649 to 2332 by at least 3 amino acid residues.

22. The covalent conjugate of claim 19, wherein said poly(ethylene glycol) is a monomethoxy-poly(ethylene glycol).

23. The covalent conjugate of claim 19, wherein said poly(ethylene glycol) is a member selected from linear poly(ethylene glycol) and branched poly(ethylene glycol).

24. The covalent conjugate of claim 19, wherein said poly(ethylene glycol) has a molecular weight distribution that is essentially homodisperse.

25. The covalent conjugate of claim 19, wherein said at least one glycosyltransferase is selected from the group consisting of ST3Gal1, ST3Gal3, ST6GalNAcI, CST-II and combinations thereof.

26. The covalent conjugate of claim 19, said method further comprising:

(b) contacting said covalent conjugate with a sialic acid donor and a sialyltransferase under conditions suitable for said sialyltransferase to transfer a sialic acid residue onto said covalent conjugate, thereby transferring a sialic acid moiety onto said covalent conjugate.

27. A covalent conjugate formed between a poly(ethylene glycol) and a Factor VIII peptide, said peptide having the formula:

28. The covalent conjugate of claim 27, wherein said Factor VIII peptide is a Factor VIII variant wherein the Factor VIII variant further comprises a B-domain deletion.

29. The covalent conjugate of claim 28, wherein said Factor VIII variant further comprises amino acids 1-740 linked to amino acids 1649 to 2332 by at least 3 amino acid residues.

30. The covalent conjugate of claim 27, said method further comprising:

(b) contacting said covalent conjugate with a sialic acid donor and a sialyltransferase under conditions suitable for said sialyltransferase to transfer a sialic acid residue onto said covalent conjugate, thereby transferring a sialic acid moiety onto said covalent conjugate.

31. The covalent conjugate according to claim 27, wherein said poly(ethylene glycol) has a molecular weight that is essentially homodisperse.

32. The covalent conjugate of claim 27, wherein said poly(ethylene glycol) is a member selected from linear poly(ethylene glycol) and branched poly(ethylene glycol).

33. The covalent conjugate of claim 27, wherein said poly(ethylene glycol) is monomethoxy-poly(ethylene glycol).

34. The covalent conjugate of claim 27, wherein said at least one glycosyltransferase is a member selected from the group consisting of GalT, ST3Gal3, CST-II and combinations thereof.

35. A covalent conjugate formed between a poly(ethylene glycol) and a Factor VIII peptide, said peptide having the formula:

36. The covalent conjugate of claim 35, wherein said Factor VIII peptide is a Factor VIII variant wherein the Factor VIII variant further comprises a B-domain deletion.

37. The covalent conjugate of claim 36, wherein said Factor VIII variant further comprises amino acids 1-740 linked to amino acids 1649 to 2332 by at least 3 amino acid residues.

38. The covalent conjugate of claim 35, said method further comprising:

(c) contacting said covalent conjugate with a sialic acid donor and a sialyltransferase under conditions suitable for said sialyltransferase to transfer a sialic acid residue onto said covalent conjugate, thereby transferring a sialic acid moiety onto said covalent conjugate.

39. The covalent conjugate according to claim 35, wherein said poly(ethylene glycol) has a molecular weight that is essentially homodisperse.

40. The covalent conjugate of claim 35, wherein said poly(ethylene glycol) is a member selected from linear poly(ethylene glycol) and branched poly(ethylene glycol).

41. The covalent conjugate of claim 35, wherein said poly(ethylene glycol) is monomethoxy-poly(ethylene glycol).

42. The covalent conjugate of claim 35, wherein said at least one glycosyltransferase is selected from the group consisting of ST3Gal3, ST3Gal1, ST6GalNAcI, CST-II and combinations thereof.

43. A covalent conjugate formed between a poly(ethylene glycol) and a Factor VIII peptide, said peptide having the formula:

44. The covalent conjugate of claim 43, wherein said Factor VIII peptide is a Factor VIII variant wherein the Factor VIII variant further comprises a B-domain deletion.

45. The covalent conjugate of claim 44, wherein said Factor VIII variant further comprises amino acids 1-740 linked to amino acids 1649 to 2332 by at least 3 amino acid residues.

46. The covalent conjugate of claim 43, wherein said method further comprises removing of step (a) produces a truncated glycan in which a, b, c, e and x are each 0.

47. The covalent conjugate of claim 43, wherein X³, X⁵, and X⁷, are selected from the group consisting of (mannose)_z and (mannose)_z-(X⁸)_y

wherein

X⁸ is a glycosyl moiety selected from mono- and oligo-saccharides;

y is an integer selected from 0 and 1; and

z is an integer between 1 and 20, wherein

when z is 3 or greater, (mannose)_z is selected from linear and branched structures.

48. The covalent conjugate of claim 43, wherein X⁴ is selected from the group consisting of GlcNAc and xylose.

49. The covalent conjugate of claim 43, wherein X³, X⁵, and X⁷ are (mannose)_u, wherein

u is selected from the integers between 1 and 20, and when u is 3 or greater, (mannose)_u is selected from linear and branched structures.

50. The covalent conjugate of claim 43, said method further comprising:

(c) contacting said covalent conjugate with a sialic acid donor and a sialyltransferase under conditions suitable for said sialyltransferase to transfer a sialic acid residue onto said covalent conjugate, thereby transferring a sialic acid moiety onto said covalent conjugate.

51. The covalent conjugate of claim 43, wherein said poly(ethylene glycol) has a molecular weight distribution that is essentially homodisperse.

52. The covalent conjugate of claim 43, wherein said poly(ethylene glycol) is a member selected from linear poly(ethylene glycol) and branched poly(ethylene glycol).

53. The covalent conjugate of claim 43, wherein said poly(ethylene glycol) is monomethoxy-poly(ethylene glycol).

54. The covalent conjugate of claim 43, wherein said at least one glycosyltransferase is selected from the group consisting of ST3Gal3, ST3Gal1, ST6GalNAcI CST-II and combinations thereof.

55. A covalent conjugate formed between a poly(ethylene glycol) and a glycosylated or non-glycosylated Factor VIII peptide, wherein said polymeric modifying group is conjugated to said Factor VIII peptide via an intact glycosyl linking group interposed between and covalently linked to both said Factor VIII peptide and said poly(ethylene glycol), wherein said covalent conjugate is prepared by a cell-free in vitro method comprising:

(a) contacting said Factor VIII peptide with a mixture comprising a nucleotide sugar covalently linked to said poly(ethylene glycol), and a glycosyltransferase for which said nucleotide sugar is a substrate under conditions suitable for said at least one glycosyltransferase to transfer a modified sugar moiety of said nucleotide sugar to said Factor VIII peptide, wherein said modified sugar moiety comprises at least one modifying group which is a poly(ethylene glycol),

thereby forming said covalent conjugate of said Factor VIII peptide.

56. The covalent conjugate of claim 55, wherein said Factor VIII peptide is a Factor VIII variant wherein the Factor VIII variant further comprises a B-domain deletion.

57. The covalent conjugate of claim 56, wherein said Factor VIII variant further comprises amino acids 1-740 linked to amino acids 1649 to 2332 by at least 3 amino acid residues.

58. The covalent conjugate of claim 55, wherein said glycosyl linking group is covalently attached to a glycosyl residue covalently attached to said peptide.

59. The covalent conjugate of claim 55, wherein said glycosyl linking group is covalently attached to an amino acid residue of said peptide.

60. The covalent conjugate of claim 55, wherein said poly(ethylene glycol) has a degree of polymerization from about 1 to about 40,000.

61. The covalent conjugate of claim 55, wherein said poly(ethylene glycol) has a degree of polymerization from about 1 to about 80,000, and further wherein said poly(ethylene glycol) is a branched structure.

62. The covalent conjugate of claim 55, wherein said poly(ethylene glycol) has a mass from about 10 kDa to about 40 kDa.

63. The covalent conjugate of claim 55, wherein said poly(ethylene glycol) has a degree of polymerization from about 1 to about 30,000, and further wherein said poly(ethylene glycol) is a linear structure.

64. The covalent conjugate of claim 60, wherein said poly(ethylene glycol) has a degree of polymerization from about 1 to about 5,000.

65. The covalent conjugate of claim 64, wherein said poly(ethylene glycol) has a degree of polymerization from about 1 to about 1,000.

66. The covalent conjugate of claim 55, wherein said poly(ethylene glycol) is monomethoxy-poly(ethylene glycol).

67. The covalent conjugate of claim 55, wherein said poly(ethylene glycol) has a molecular weight distribution that is essentially homodisperse.

68. The covalent conjugate of claim 55, wherein said poly(ethylene glycol) is a member selected from linear poly(ethylene glycol) and branched poly(ethylene glycol).

69. The covalent conjugate of claim 55, wherein said glycosyltransferase is selected from the group consisting of ST3Gal3, ST3Gal1, ST6GalNAcI, CST-II and combinations thereof.

70. The covalent conjugate of claim 55, said method further comprising:

(b) contacting said covalent conjugate with a sialic acid donor and a sialyltransferase under conditions suitable for said sialyltransferase to transfer a sialic acid residue onto said covalent conjugate, thereby transferring a sialic acid moiety onto said covalent conjugate.

71. The covalent conjugate of claim 55, wherein said glycosyltransferase is selected from the group consisting of sialyltransferase, galactosyltransferase, glucosyltransferase, GalNAc transferase, GlcNAc transferase, fucosyltransferase, mannosyltransferase and combinations thereof.

72. The covalent conjugate of claim 55, wherein said glycosyltransferase is recombinantly produced.

73. The covalent conjugate of claim 72, wherein said glycosyltransferase is a recombinant prokaryotic enzyme.

74. The covalent conjugate of claim 72, wherein said glycosyltransferase is a recombinant eukaryotic enzyme.

75. The covalent conjugate of claim 55, wherein said glycosylated peptide is partially deglycosylated prior to said contacting.

76. The covalent conjugate of claim 55, wherein said intact glycosyl linking group is a sialic acid residue.

77. The covalent conjugate of claim 55, wherein said method is performed in a cell-free environment.

78. The covalent conjugate of claim 55, wherein said covalent conjugate is isolated.

79. The covalent conjugate of claim 78, wherein said covalent conjugate is isolated by membrane filtration.

80. A covalent conjugate formed between a poly(ethylene glycol) and a Factor VIII peptide, said peptide having the formula:

81. The covalent conjugate of claim 80, wherein said Factor VIII peptide is a Factor VIII variant wherein the Factor VIII variant further comprises a B-domain deletion.

82. The covalent conjugate of claim 81, wherein said Factor VIII variant further comprises amino acids 1-740 linked to amino acids 1649 to 2332 by at least 3 amino acid residues.

83. The covalent conjugate of claim 80, said method further comprising:

(b) contacting said covalent conjugate with a sialic acid donor and a sialyltransferase under conditions suitable for said sialyltransferase to transfer a sialic acid residue onto said covalent conjugate, thereby transferring a sialic acid moiety onto said covalent conjugate.

84. The covalent conjugate according to claim 80, wherein said poly(ethylene glycol) has a molecular weight that is essentially homodisperse.

85. The covalent conjugate of claim 80, wherein said poly(ethylene glycol) is a member selected from linear poly(ethylene glycol) and branched poly(ethylene glycol).

86. The covalent conjugate of claim 80, wherein said poly(ethylene glycol) is monomethoxy-poly(ethylene glycol).

87. The covalent conjugate of claim 80, wherein said at least one glycosyltransferase is selected from the group consisting of GalNAcT2, ST3Gal1, ST3Gal3, ST6GalNAcI, CST-I, CST-II and combinations thereof.

A covalent conjugate formed between a Factor VIII peptide and a poly(ethylene glycol), wherein said poly(ethylene glycol) is covalently attached to said Factor VIII peptide through an intact glycosyl linking group, said Factor VIII peptide comprising a glycosyl residue having a formula which is a member selected from:

89. The covalent conjugate of claim 88, wherein said Factor VIII peptide is a Factor VIII variant wherein the Factor VIII variant further comprises a B-domain deletion.

90. The covalent conjugate of claim 89, wherein said Factor VIII variant further comprises amino acids 1-740 linked to amino acids 1649 to 2332 by at least 3 amino acid residues.

91. The covalent conjugate of claim 88, said method further comprising:

(b) contacting said covalent conjugate with a sialic acid donor and a sialyltransferase under conditions suitable for said sialyltransferase to transfer a sialic acid residue onto said covalent conjugate, thereby transferring a sialic acid moiety onto said covalent conjugate.

92. The covalent conjugate according to claim 88, wherein said poly(ethylene glycol) has a molecular weight that is essentially homodisperse.

93. The covalent conjugate of claim 88, wherein said poly(ethylene glycol) is a member selected from linear poly(ethylene glycol) and branched poly(ethylene glycol).

94. The covalent conjugate of claim 88, wherein said poly(ethylene glycol) is monomethoxy-poly(ethylene glycol).

95. The covalent conjugate of claim 88, wherein said at least one glycosyltransferase is selected from the group consisting of ST3Gal3, CST-II and combinations thereof.

96. The covalent conjugate of claim 88, said method further comprising:

(c) prior to step (a), contacting said Factor VIII peptide with a sialidase under conditions appropriate to remove a sialic acid from said Factor VIII peptide, thereby removing said sialic acid from said Factor VIII peptide.

97. The covalent conjugate of claim 88, said method further comprising:

(d) contacting the product of step (a) with a sialyltransferase and a sialic acid donor under conditions appropriate to transfer a sialic acid to said product, thereby transferring said sialic acid to said product.

98. The covalent conjugate of claim 88, said method further comprising:

(e) prior to step (a), contacting said Factor VIII peptide with a galactosyl transferase and a galactose donor under conditions appropriate to transfer a galactose to said Factor VIII peptide, thereby transferring said galactose to said Factor VIII peptide.

99. The covalent conjugate of claim 88, said method further comprising:

(f) contacting the product from step (a) with a moiety that reacts with said poly(ethylene glycol), thereby forming a conjugate between said intact glycosyl linking group and said poly(ethylene glycol).

100. The covalent conjugate of claim 88, said method further comprising:

(g) prior to step (a), contacting said Factor VIII peptide with N-acetylglucosamine transferase and a GlcNAc donor under conditions appropriate to transfer a GlcNAc to said Factor VIII peptide, thereby transferring said GlcNAc to said Factor VIII peptide.

101. The covalent conjugate of claim 88, said method further comprising:

(h) prior to step (a), contacting said Factor VIII peptide with a sialidase under conditions appropriate to remove a sialic acid from said Factor VIII peptide, thereby removing said sialic acid from Factor VIII peptide.

102. The covalent conjugate of claim 88, said method further comprising:

(i) prior to step (a), contacting said Factor VIII peptide with N-acetylglucosamine transferase and a GlcNAc donor under conditions appropriate to transfer a GlcNAc to said Factor VIII peptide, thereby transferring said GlcNAc to said Factor VIII peptide.

The covalent conjugate of claim 100, said method further comprising contacting said Factor VIII peptide with galactosyl transferase and a galactose donor under conditions appropriate to transfer a galactose to said Factor VIII peptide, thereby transferring said galactose to said Factor VIII peptide.

104. The covalent conjugate of claim 88, said method further comprising:

(j) prior to step (a), contacting said Factor VIII peptide with endoglycanase under conditions appropriate to cleave a glycosyl moiety from said Factor VIII peptide, thereby cleaving said glycosyl moiety from said Factor VIII peptide.

The covalent conjugate of claim 104, said method further comprising contacting said Factor VIII peptide with galactosyl transferase and a galactose donor under conditions appropriate to transfer a galactose to said Factor VIII peptide, thereby transferring said galactose to said Factor VIII peptide.

106. The covalent conjugate of claim 88, said method further comprising:

(k) prior to step (a), contacting said Factor VIII peptide with ST3Gal3 and a sialic acid donor under conditions appropriate to transfer a sialic acid to said product, thereby transferring said sialic acid to said product.

107. The covalent conjugate of claim 88, said method further comprising:

(l) prior to step (a), contacting said Factor VIII peptide with a mannosidase under conditions appropriate to remove a mannose from said Factor VIII peptide, thereby removing said mannose from said Factor VIII peptide.

The covalent conjugate of claim 107, said method further comprising contacting said Factor VIII peptide with N-acetylglucosamine transferase and a GlcNAc donor under conditions appropriate to transfer a GlcNAc to said Factor VIII peptide, thereby transferring said GlcNAc to said Factor VIII peptide.

109. The covalent conjugate of claim 108, said method further comprising contacting said Factor VIII peptide with galactosyl transferase and a galactose donor under conditions appropriate to transfer a galactose to said Factor VIII peptide, thereby transferring said galactose to said Factor VIII peptide.

110. The covalent conjugate of claim 88, wherein

e, f, g, and h are members independently selected from the integers between 1 and 4;

a, b, c, d, i, j, k, l, m, n, o, p, q, r, s, t, u, aa, and cc are members independently selected from 0 and 1; and

v, w, x, y, z, and dd are 0.
