Abstract

A composite biocompatible hydrogel material includes a porous polymer matrix, the polymer matrix including a plurality of pores and providing a Young'"'"'s modulus of at least 10 GPa. A calcium comprising salt is disposed in at least some of the pores. The porous polymer matrix can comprise cellulose, including bacterial cellulose. The composite can be used as a bone graft material. A method of tissue repair within the body of animals includes the steps of providing a composite biocompatible hydrogel material including a porous polymer matrix, the polymer matrix including a plurality of pores and providing a Young'"'"'s modulus of at least 10 GPa, and inserting the hydrogel material into cartilage or bone tissue of an animal, wherein the hydrogel material supports cell colonization in vitro for autologous cell seeding.

Claims

1. A composite material, made by a process comprising providing bacterial cellulose which includes a plurality of pores, immersing said bacterial cellulose in a solution containing a calcium source, and immersing said bacterial cellulose in a separate solution containing a source of phosphate, thereby forming said composite material which consists essentially of bacterial cellulose and calcium deficient hydroxyapatite, said calcium deficient hydroxyapatite being in the form of crystals disposed in said plurality of pores.

3. The composite material of claim 1 or 2, wherein said calcium source is at least one selected from the group consisting of calcium phosphate, calcium chloride, calcium fluoride, calcium carbonate, calcium hydroxide, and calcium nitrate.

4. The composite material of claim 1 or 2, wherein said source of phosphate is at least one selected from the group consisting of sodium phosphate dibasic, potassium phosphate dibasic, sodium tripolyphosphate, and diammonium phosphate.

5. The composite material of claim 1 or 2, wherein the immersing in said solution containing a calcium source is performed before the immersing in said solution containing a source of phosphate.

6. The composite material of claim 1 or 2, wherein the immersing in said solution containing a calcium source is performed after the immersing in said solution containing a source of phosphate.

7. The composite material of claim 1 or 2, wherein the immersing in said solution containing a calcium source and the immersing in said solution containing a source of phosphate are performed in multiple cycles.

8. The composite material of claim 1 or 2, wherein said bacterial cellulose provides a Young'"'"'s modulus of at least 10 GPa.

9. The composite material of claim 6, wherein said solution containing a source of phosphate is a solution containing phosphoric acid and urea, and the bacterial cellulose becomes phosphorylated after immersing in said solution containing a source of phosphate.

2. A composite material, made by a process comprising providing bacterial cellulose which includes a plurality of pores, immersing said bacterial cellulose in a solution containing a calcium source, and immersing said bacterial cellulose in a separate solution containing a source of phosphate, thereby forming said composite material which consists essentially of bacterial cellulose, calcium deficient hydroxyapatite, and at least one of bone morphogenetic proteins (BMP'"'"'s) or chondrocytes, said calcium deficient hydroxyapatite being in the form of crystals disposed in said plurality of pores.
