Abstract

A wound closure apparatus is provided which utilizes blood fluid by activating the clotting cascade of blood fluid outside the body within a substantially enclosed sterile container then introducing, the blood fluid to the wound site to complete clotting. An apparatus for providing ways of inhibiting anticoagulating agents, and slowing fibrin clot degradation are also disclosed. Kits for practicing the invention singularly or in combination with, and/or associated with preferred procedures are also disclosed. The invention provides a clotting cascade initiation apparatus (1) including a substantially enclosed sterile containment chamber within which an aliquot of blood fluid, either autologous or from donor sources can be received, and retained. In preferred embodiments, the sterile containment chamber further includes a heparin binding agent which will bind heparin and remove it from the blood fluid. In further embodiments, the containment chamber will also include a procoagulating agent, wherein a clotting cascade can be initiated when the blood fluid is accepted into the sterile containment chamber.

Claims

1. A coating for a surface comprising:

at least a first coating, a second coating, and a third coating, wherein the first coating is in contact with at least a portion of the surface and the second coating is disposed between the first and the third coating, with the second coating comprising a high molecular weight negatively charged polymer,

wherein the first coating, the third coating, or both the first coating and the third coating comprises water soluble butylated polyethyleneimine and

a fourth coating of heparin on the third coating.

2. The coating of claim 1, wherein the high molecular weight negatively charged polymer is polyacrylic acid with a molecular weight of at least 3,000,000.

3. The coating of claim 1, wherein the second coating is a coating made from a 0.06% w/v solution of polyacrylic acid in aqueous solution.

The coating of claim 1 wherein the third coating comprises polyethyleneimine crosslinked with a member of the group consisting of crotenaldheyde, carbonyl diimidazole, ethylene glycol diacrylate, butane diol diglycidyl ether, pentaerythzito-tris-(N-aridnyl) proprionate, and polyethylene oxide diglycidyl ether.

5. The coating of claim 4, wherein the negatively charged high molecular weight polymer is polyacrylic acid with a molecular weight of at least 3,000,000.

6. The coating of claim 4, wherein the second coating is a coating made from a 0.06% w/v solution of polyacrylic acid in aqueous solution.

7. The coating of claim 4, wherein the surface is a porous matrix.

8. The coating of claim 7, wherein the surface is a plurality of glass beads.

9. The coating of claim 4, wherein the first layer and the surface are identical and the first layer comprises positively charged functional groups.

10. The coating of claim 1 wherein the first coating is the butylated polyethyleneimine, the second coating is polyacrylic acid, and the third coating is polyethyleneimine.

11. The coating of claim 10 wherein the polyacrylic acid has a molecular weight of at least three million.

12. The coating of claim 1, wherein the surface is a porous matrix.

13. The coating of claim 12, wherein the surface is a plurality of glass beads.

14. A coating for a surface comprising:

at least a first coating, a second coating, and a third coating, wherein the first coating is in contact with at least a portion of the surface and the second coating is disposed between the first and the third coating,

the first and third coatings comprising polyethyleneimine and

the second coating comprising polyacrylic acid with a molecular weight of at least 3,000,000,

and a fourth coating on the third coating that comprises heparin.

15. The coating of claim 14, wherein the first coating and/or the third coating comprises butylated polyethyleneimine.

16. A method for coating a surface comprising:

coating at least a portion of the surface with a first coating of a positively charged polymer;

coating at least a portion of the first coating with a second coating that contacts the first coating, wherein the second coating includes a high molecular weight negatively charged polymer; and

coating at least a portion of the second coating with a third coating of a positively charged polymer that contacts the second coating,

wherein the positively charged polymer in the first coating, the third coating, or both the first coating and the third coating comprises water soluble butylated polyethyleneimine,

and further comprising applying a layer of heparin to the third coating by exposing the third coating to a solution that comprises the heparin.

17. The method of claim 16, wherein the third coatings comprise polyethyleneimine.

18. The method of claim 16, wherein the high molecular weight negatively charged polymer comprises polyacrylic acid having a molecular weight of at least 3,000,000.

19. The method of claim 18, wherein the second coating is a coating made from a 0.06% w/v solution of polyacrylic acid in aqueous solution.

20. The method of claim 16 wherein the first coating is the water soluble butylated polyethyleneimine, the second coating is polyacrylic acid, and the third coating is polyethyleneimine.

21. The method of claim 20 wherein the polyacrylic acid has a molecular weight of at least 3,000,000.

22. The method of claim 16 wherein the surface is a porous matrix.

23. The method of claim 22 wherein the surface is a plurality of glass beads.
