Abstract

The present invention relates to one or more peptides which may be used to test for the presence of antibodies to B. microti. These peptide are selected based on the strength and specificity of the human antibody response to B. microti. These peptides alone or in combination have a variety of uses, for example, as a component of a diagnostic assay or kit.

Claims

1. An agent selected from the group consisting of the following:

(a) a first peptide and an optional second peptide, wherein said first peptide consists of a sequence as set forth in SEQ ID NO;

7;

(b) a chimeric protein consisting of said first peptide and a second peptide; and

(c) a fusion protein consisting of a first portion and a second portion, wherein the first portion is a protein according to

(b) and the second portion is a binding member.

4. A method of detecting the presence of antibodies to B. microti in a test sample comprising the steps of

(a) contacting said test sample with an agent according to claim 1 for a time and under conditions sufficient to form agent/antibody complexes;

and

(b) detecting the presence of said agent/antibody complexes, presence of said complexes indicating presence of antibodies to B. microti in said test sample.

5. The method of claim 4, wherein said test sample is selected from the group consisting of blood, serum, plasma, saliva, cerebrospinal fluid and urine.

6. The method of claim 4, wherein the agent is attached to a solid support.

7. A method of diagnosing B. microti injection in a subject comprising the steps of:

(a) detecting the presence of antibodies to B. microti in a test sample according to the method of claim 4; and

(b) comparing the amount of anti-B. microti antibody to a control, wherein a level of anti-B. microti antibody above the control is indicative of B. microti infection.

8. A method of detecting antibodies to B. microti in a test sample comprising the steps of:

(a) contacting said test sample with an agent according to claim 1 for a time and under conditions sufficient to form agent/antibody complexes;

(b) adding a conjugate to the resulting agent/antibody complexes for a time and under conditions sufficient to allow said conjugate to bind to the bound antibody, wherein said conjugate comprises an anti-antibody attached to a signal-generating compound capable of generating a detectable signal; and

(c) detecting presence of antibody which may be present in said sample by detecting a signal generated by said signal-generating compound, presence of said signal indicating the presence of antibodies to B. microti in said test sample.

9. The method of claim 8, wherein said test sample is selected from the group consisting of blood, serum, plasma, saliva, cerebrospinal fluid and urine.

10. The method of claim 8, wherein the agent is attached to a solid support.

11. A method of diagnosing B. microti injection in a subject comprising the steps of:

(a) detecting the presence of antibodies to B. microti in a test sample according to the method of claim 8; and

(b) comparing the amount of anti-B. microti antibody to a control, wherein a level of anti-B microti antibody above the control is indicative of B. microti infection in said subject.

2. An agent comprising a chimeric protein, wherein said chimeric protein comprises a first peptide and a second peptide, and wherein said chimeric protein comprises the sequence of SEQ ID NO:

An agent comprising a fusion protein, wherein said fusion protein comprises a first and a second portion, wherein said first portion is a chimeric protein comprising a first peptide and a second peptide and said second portion is a binding member, and wherein said fusion protein comprises the sequence of SEQ ID NO:
