1. A method of processing a sample in a receptacle having a plurality of interconnected chambers, including a detection chamber, the method comprising the steps of:

  • (a) moving a compression element positioned between the receptacle and a detector into engagement with a flexible portion of the detection chamber, thereby depressing the flexible portion to agitate the contents of the detection chamber or to move the contents of the detection chamber into a directly connected chamber of the receptacle, wherein an actuator moves the compression element relative to the detector;

    (b) transmitting excitation light from a light source of the detector through the compression element and to the detection chamber; and

    (c) detecting a light signal transmitted through the compression element from the contents of the detection chamber with a signal detecting element of the detector.
