1. A method for determining the amount of thyroglobulin in a test sample, comprising:

  • (a) digesting thyroglobulin from the test sample to form peptide T129;

    (b) ionizing the peptide T129 to generate a multiply charged ion detectable by mass spectrometry;

    wherein the ion has a mass/charge ratio (m/z) of 636.4±

    0.5; and

    (c) detecting the amount of one or more T129 fragment ions each having a m/z selected from the group consisting of 541.3±

    0.5, 612.3±

    0.5, 636.4±

    0.5, 726.4±

    0.5, 797.4±

    0.5, 912.4±

    0.5, and 1059.5±

    0.5 by mass spectrometry;

    wherein the amount of the ions detected is related to the amount of the thyroglobulin in the test sample.
