Abstract

The present invention relates to control of pest infestation by inhibiting one or more biological functions. The invention provides methods and compositions for such control, By feeding one or more recombinant double stranded RNA molecules provided by the invention to the pest, a reduction in pest infestation is obtained through suppression of gene expression. The invention is also directed to methods for making transgenic plants that express the double stranded RNA molecules, and to particular combinations of transgenic pesticidal agents for use in protecting plants from pest infestation.

Claims

1. A nucleic acid molecule comprising a recombinant polynucleotide selected from the group consisting of:

(a) a polynucleotide comprising the nucleic acid sequence of SEQ ID NO;

(b) a polynucleotide comprising at least 95% sequence identity to SEQ ID NO;

841;

(c) a fragment of at least 23 contiguous nucleotides of a nucleic acid sequence of SEQ ID NO;

841, wherein ingestion by a corn rootworm of a double stranded ribonucleotide sequence comprising at least one strand that is complementary to said fragment inhibits the growth of said corn rootworm; and

(d) a complement of the sequence of (a), (b), or (c);

wherein said nucleic acid molecule further comprises a heterologous promoter operably linked to said recombinant polynucleotide.

2. A plant transformation vector comprising the nucleic acid of claim 1.

3. A double stranded ribonucleotide hairpin molecule produced from the expression of the polynucleotide according to claim 1, wherein ingestion of said ribonucleotide hairpin molecule by a corn rootworm inhibits the growth of said corn rootworm.

The double stranded ribonucleotide hairpin molecule of claim 3, defined as produced by preparing a recombinant polynucleotide sequence comprising a first, a second and a third polynucleotide sequence, wherein the first polynucleotide sequence comprises the recombinant polynucleotide of claim 1, wherein the third polynucleotide sequence is linked to the first polynucleotide sequence by the second polynucleotide sequence, and wherein the third polynucleotide sequence is substantially the reverse complement of the first polynucleotide sequence such that the first and the third polynucleotide sequences hybridize when transcribed into a ribonucleic acid to form the double stranded ribonucleotide hairpin molecule stabilized by the linked second ribonucleotide sequence.

5. The double stranded ribonucleotide hairpin molecule of claim 3, wherein ingestion of the ribonucleotide hairpin molecule by the corn rootworm inhibits the expression of a nucleotide sequence complementary to said polynucleotide sequence.

6. A cell transformed with the nucleic acid molecule of claim 1.

7. The cell of claim 6, defined as a prokaryotic cell.

8. The cell of claim 6, defined as a eukaryotic cell.

9. The cell of claim 6, defined as a plant or bacterial cell.

10. A plant transformed with the nucleic acid molecule of claim 1.

11. A seed of the plant of claim 10, wherein the seed comprises the nucleic acid molecule.

12. The plant of claim 10, wherein said polynucleotide is expressed in a cell of the plant as a double stranded ribonucleotide hairpin molecule and ingestion of a corn rootworm inhibitory amount of said double stranded ribonucleotide hairpin molecule in a diet inhibits the corn rootworm from further feeding on said diet.

13. The plant of claim 12, wherein the corn rootworm is selected from the group consisting of Diabrotica virgifera, Diabrotica virgifera virgifera, Diabrotica virgifera zea, Diabrotica balteata, Diabrotica barberi, Diabrotica viridula, Diabrotica speciosa, and Diabrotica undecimpunctata.

14. The plant of claim 12, wherein ingestion of the com rootworm inhibitory amount of the double stranded ribonucleotide hairpin molecule stunts the growth of the com rootworm.

15. A commodity product produced from a plant according to claim 10, wherein said commodity product comprises a detectable amount of the polynucleotide of claim 1 or a ribonucleotide expressed therefrom.

18. A method for controlling a corn rootworm infestation comprising providing in the diet of a corn rootworm a plant cell expressing a polynucleotide sequence according to claim 1, wherein the polynucleotide is expressed to produce a double stranded ribonucleic acid that functions upon ingestion by the corn rootworm to inhibit the expression of a target sequence within said corn rootworm and results in decreased feeding on said diet relative to a diet lacking the plant cell.

19. The method of claim 18, wherein the corn rootworm exhibits reduced growth following ingestion of the cell.

The method of claim 18, wherein the plant cell further comprises a polynucleotide sequence encoding a pesticidal agent selected from the group consisting of a patatin, a Bacillus thuringiensis insecticidal protein, a Xenorhabdus insecticidal protein, a Photorhabdus insecticidal protein, a Bacillus laterosporus insecticidal protein, and a Bacillus sphaericus insecticidal protein.

The method of claim 20 wherein said Bacillus thuringiensis insecticidal protein is selected from the group consisting of a Cry1, a Cry3, a TIC851, a CryET70, a Cry22, a TIC901, a TIC201, a TIC407, a TIC417, a binary insecticidal protein CryET33 and CryET34, a binary insecticidal protein CryET80 and CryET76, a binary insecticidal protein TIC 100 and TIC101, a combination of the insecticidal proteins ET29 or ET37 with insecticidal proteins TIC810 or TIC812, and a binary insecticidal protein PS149B1.

The method of claim 18, wherein said corn rootworm is a Diabrotica spp. selected from the group consisting of Diabrotica virgifera, Diabrotica virgifera virgifera, Diabrotica virgifera zea, Diabrotica balteata, Diabrotica barberi, Diabrotica viridula, Diabrotica speciosa, and Diabrotica undecimpunctata.

23. A method for improving the yield of a crop produced from a crop plant subjected to corn rootworm infestation, said method comprising the steps of, a) introducing a nucleic acid molecule according to claim 1 into said crop plant, b) cultivating the crop plant to allow the expression of said polynucleotide, wherein expression of the polynucleotide inhibits feeding by corn rootworms and loss of yield due to corn rootworm infestation.

The method of claim 23, wherein the corn rootworm pest is selected from the group consisting of Diabrotica undecimpunctata howardi (Southern Corn Rootworm (SCR)), Diabrotica virgifera virgifera (Western Corn Rootworm (WCR)), Diabrotica barberi (Northern Corn Rootworm (NCR)), Diabrotica virgifera zea (Mexican Corn Rootworm (MCR)), Diabrotica balteata (Brazilian Corn Rootworm (BZR)), Diabrotica viridula (Brazilian Corn Rootworm (BZR)), and Diabrotica speciosa (Brazilian Corn Rootworm (BZR)).

25. A method for improving the drought tolerance of a crop produced from a crop plant subjected to corn rootworm infestation, said method comprising the steps of, a) introducing a nucleic acid molecule according to claim 1 into said crop plant, b) cultivating the crop plant to allow the expression of said polynucleotide, wherein expression of the polynucleotide inhibits feeding by corn rootworms and loss of drought tolerance due to corn rootworm infestation.

26. A method of producing a commodity product comprising obtaining a plant according to claim 10 or a part thereof, and preparing a commodity product from the plant or part thereof.

27. A method of producing food or feed, comprising obtaining a plant according to claim 10 or a part thereof and preparing food or feed from said plant or part thereof.

28. The method of claim 27, wherein the food or feed is defined as oil, meal, protein, starch, flour or silage.

29. The plant of claim 10, wherein said plant is a corn plant.

30. The seed of claim 11, wherein said seed is corn seed.

31. The cell according to claim 9, wherein said plant cell is a corn plant cell.

32. A cell of the plant of claim 29, wherein said cell comprises the nucleic acid molecule.

16. A method for controlling corn rootworm infestation comprising providing in the diet of a corn rootworm an agent comprising a first polynucleotide sequence that functions upon ingestion by the corn rootworm to inhibit a biological function within said corn rootworm, wherein said polynucleotide sequence exhibits from about 95 to about 100% nucleotide sequence identity along at least from about 19 to about 25 contiguous nucleotides to a coding sequence derived from said corn rootworm and is hybridized to a second polynucleotide sequence that is complementary to said first polynucleotide sequence, and wherein said coding sequence derived from said corn rootworm is selected from the group consisting of SEQ ID NO:

841, and the complement thereof.

The method of claim 16, wherein said corn rootworm is a Diabrotica spp. selected from the group consisting of Diabrotica virgifera, Diabrotica virgifera virgifera, Diabrotica virgifera zea, Diabrotica balteata, Diabrotica barberi, Diabrotica viridula, Diabrotica speciosa, and Diabrotica undecimpunctata.
