A method of inhibiting at least one of unwanted growth, proliferation or survival of a cell, comprising contacting said cell with an effective amount of a hedgehog RNAi antagonist against a Sonic hedgehog (Shh) gene, wherein said RNAi antagonist comprises a sequence that is at least 90% identical to a nucleic acid sequence set forth in any of SEQ ID NOs:
- 5, 6, 9, or 10, wherein said RNAi antagonist is 19-30 nucleotides in length, wherein contacting said cell with said hedgehog RNAi antagonist decreases at least one of cell growth, proliferation or survival, wherein said cell is a cancer cell, and wherein said cancer cell is a colon cancer cell.

Abstract
The instant application relates to methods and reagents for modulating the Hedgehog signaling pathway using RNA interference technology (RNAi). The application provides potential targets of the Hedgehog RNAi, methods to identify additional Hedgehog signaling pathway components, methods to inhibit Hedgehog signaling targets using siRNA, and their uses in the treatment of a number of disease conditions.
