Abstract

The present invention provides compositions comprising human placental telopeptide collagen, methods of preparing the compositions, methods of their use and kits comprising the compositions. The compositions, kits and methods are useful, for example, for augmenting or replacing tissue of a mammal.

Claims

1. A method of making a composition comprising telopeptide collagen, comprising, in order, the steps of:

(a) macerating placental tissue;

(b) suspending the placental tissue in a hypertonic saline solution;

(c) suspending the placental tissue in water; and

(d) treating the placental tissue with a mild detergent,

wherein said collagen has not been chemically modified or contacted with a base as part of the method, and wherein said composition additionally comprises 10% to about 12% elastin by dry weight, glycosaminoglycans, and less than 1% fibronectin or less than 1% laminin by weight after said method is completed.

2. The method of claim 1, wherein the hypertonic saline solution comprises sodium chloride.

3. The method of claim 1, further comprising the step of filtering the placental tissue.

4. The method of claim 1, wherein said placental tissue is human placental tissue.

9. The method of claim 1, wherein said composition comprises between 74% and 92% Type I collagen by weight.

10. The method of claim 9, wherein said composition further comprises 4% to 6% Type III collagen or 2% to 15% Type IV collagen by weight.

11. The method of claim 1, wherein said detergent is deoxycholate.

15. The method of claim 1, wherein said composition comprises about 12% elastin.

5. A method of making a composition comprising telopeptide collagen, comprising, in order, the steps of:

(a) macerating placental tissue;

(b) treating the placental tissue with a mild detergent, and

(c) treating the placental tissue with a base,

wherein said collagen has not been chemically modified as part of the method, and wherein said composition additionally comprises between 3% and 5% elastin by weight, glycosaminoglycans, and less than 0.01% laminin or less than 0.01% fibronectin by weight.

6. The method of claim 5, wherein said base comprises ammonium hydroxide, potassium hydroxide, or sodium hydroxide.

7. The method of claim 5, further comprising the step of filtering the placental tissue.

8. The method of claim 5, wherein said placental tissue is human placental tissue.

12. The method of claim 5, wherein said composition comprises between 74% and 92% Type I collagen by weight.

13. The method of claim 12, wherein said composition further comprises 4% to 6% Type III collagen or 2% to 15% type IV collagen by weight.

14. The method of claim 5, wherein said detergent is deoxycholate.
