Abstract

A nanofluidic channel fabricated in fused silica with an approximately 500 nm square cross section was used to isolate, detect and identify individual quantum dot conjugates. The channel enables the rapid detection of every fluorescent entity in solution. A laser of selected wavelength was used to excite multiple species of quantum dots and organic molecules, and the emission spectra were resolved without significant signal rejection. Quantum dots were then conjugated with organic molecules and detected to demonstrate efficient multicolor detection. PCH was used to analyze coincident detection and to characterize the degree of binding. The use of a small fluidic channel to detect quantum dots as fluorescent labels was shown to be an efficient technique for multiplexed single molecule studies. Detection of single molecule binding events has a variety of applications including high throughput immunoassays.

Claims

1. A device comprising:

a channel having a narrow detection portion sized to promote single molecule flow through the channel;

a light source to focus light substantially uniformly across a width of the detection portion;

means for moving fluorescent entities through the detection portion; and

a detector for detecting photon emissions from the fluorescent entities.

2. The device of claim 1 wherein the channel is approximately 500 nm deep and approximately 500 nm wide.

3. The device of claim 2 wherein the channel is formed of mirror-polished fused silica.

4. The device of claim 1 wherein the detector comprises an inverted confocal microscope for collecting emissions from the photon emitting entities.

5. The device of claim 1 wherein the fluorescent entities include organic fluorophores.

6. The device of claim 1 wherein the fluorescent entities include DNA molecules.

7. The device of claim 1 wherein the fluorescent entities include fluorescent molecules.

8. The device of claim 1 wherein the fluorescent entities include conjugated quantum dots.

9. The device of claim 8 wherein the quantum dots comprise Qdot 565 and Qdot 655.

10. The device of claim 1 wherein there is substantially no emission spectral overlap between different types of fluorescent entities.

11. The device of claim 1 wherein the fluorescent entities comprise fluorophores or proteins.

12. The device of claim 1 wherein the light source includes a laser.

13. A device comprising:

a channel having a nano-channel detection portion for single molecule flow;

a light source that focuses light substantially uniformly across a width of the detection portion;

a driver that moves fluorescen entities through the detection portion; and

a detector that detects emissions from the fluorescent entities.

14. The device of claim 13 wherein the channel is formed in a substrate and covered with a wafer.

15. The device of claim 14 wherein the substrate and wafer comprise fused silica.

16. The device of claim 15 wherein the fused silica is mirror polished.

17. The device of claim 13 wherein the channel is adapted to constrain the photon emitting entities in a lateral and axial direction with respect to the detector.

18. The device of claim 13 wherein the detector has a confined depth of focus in the channel to limit detection of unwanted emissions.

19. The device of claim 18 wherein the detector comprises an inverted confocal micro scope.

20. The device of claim 13 wherein the fluorescent entities include organic fluorophores.

21. The device of claim 13 wherein the fluorescent entities include DNA molecules.

22. The device of claim 13 wherein the fluorescent entities include fluorescent molecules.

23. The device of claim 13 wherein the fluorescent entities include conjugated quantum dots.

24. The device of claim 13 wherein the fluorescent entities have distinct emission spectra.

25. The device of claim 13 wherein the light source includes a laser.

26. A device comprising:

an array of channels, each having a nano-channel detection portion for single molecule flow, wherein the channels are formed on a substrate and covered with a wafer;

a light source that focuses light substantially uniformly across a width of the detection portion of the channels;

a driver that moves fluorescent entities through the detection portions; and

a detector that detects emissions from the fluorescent entities.

27. The device of claim 26 wherein the substrate and wafer comprise fused silica.

28. The device of claim 27 wherein the fused silica is mirror polished.

29. The device of claim 26 wherein the channel is adapted to constrain the fluorescent entities in a lateral and axial direction with respect to the detector.

30. The device of claim 26 wherein the detector has a confined depth of focus in the channel to limit detection of unwanted emissions.

31. The device of claim 30 wherein the detector comprises an inverted confocal micro scope.

32. The device of claim 26 wherein the fluorescent entities include organic fluorophores.

33. The device of claim 26 wherein the fluorescent entities include DNA molecules.

34. The device of claim 26 wherein the fluorescent entities include fluorescent molecules.

35. The device of claim 26 wherein the fluorescent entities include conjugated quantum dots.

36. The device of claim 26 wherein the fluorescent entities have distinct emission spectra.

37. A device comprising:

a channel having a nano-channel detection portion;

a light source that focuses light substantially uniformly across a width of the detection portion;

a driver that moves fluorescent entities through the detection portion; and

a detector that detects emissions from the fluorescent entities.

38. The device of claim 37 wherein the fluorescent entities include submicrometer sized nanoparticles.
