Abstract

The present invention relates generally to compositions and methods for treating neurodegenerative diseases and disorders, particularly ophthalmic diseases and disorders. Provided herein are alkoxyl derivative compounds and pharmaceutical compositions comprising these compounds. The subject compositions are useful for treating and preventing ophthalmic diseases and disorders, including age-related macular degeneration (AMD) and Stargardt'"'"'s Disease.

Claims

1. A compound of Formula (A) or tautomer, stereoisomer, or a pharmaceutically acceptable salt, thereof:

R³ and R⁴ are hydrogen;

R⁹ and R¹⁰ are each independently selected from hydrogen, halogen, C₁-C₅ alkyl optionally substituted with hydroxy or halogen, or —

or R⁹ and R¹⁰ form an oxo.

3. The compound of claim 2, or a pharmaceutically acceptable salt thereof, having the structure of Formula (C):

4. The compound of claim 3, or a pharmaceutically acceptable salt thereof, wherein n is 0 and each of R¹¹ and R¹² is hydrogen.

5. The compound of claim 4, or a pharmaceutically acceptable salt thereof, wherein, each of R³, R⁴, R¹⁴ and R¹⁵ is hydrogen.

6. The compound of claim 5, or a pharmaceutically acceptable salt thereof, wherein,

R¹ and R² are each independently selected from hydrogen, halogen, C₁-C₅ alkyl, or —

OR⁶;

R⁹ and R¹⁰ are each independently selected from hydrogen, halogen, C₁-C₅ alkyl optionally substituted with hydroxy or halogen, —

OR¹⁹;

or R⁹ and R¹⁰ together form an oxo;

R¹⁶ and R¹⁷, together with the carbon to which they are attached form a carbocyclyl; and

R¹⁸ is selected from a hydrogen, alkoxy or hydroxy.

7. The compound of claim 6, or a pharmaceutically acceptable salt thereof, wherein R¹⁶ and R¹⁷, together with the carbon to which they are attached, form a cyclohexyl or cycloheptyl and R¹⁸ is hydrogen or hydroxy.

8. The compound of claim 6, or a pharmaceutically acceptable salt thereof, wherein R¹⁶ and R¹⁷, together with the carbon to which they are attached, form a cyclobutyl, cyclopentyl, cyclohexyl, cycloheptyl, or cyclooctyl, and R¹⁸ is hydrogen or hydroxy.

9. The compound of claim 5, or a pharmaceutically acceptable salt thereof, wherein,

R¹ and R² are each independently selected from hydrogen, halogen, C₁-C₅ alkyl or —

OR⁶;

R¹⁶ and R¹⁷ is independently selected from C₁-C₁₃ alkyl; and

R¹⁸ is hydrogen, hydroxy or alkoxy.

10. The compound of claim 9, or a pharmaceutically acceptable salt thereof, wherein R¹⁶ and R¹⁷, together with the carbon atom to which they are attached form cyclopentyl, cyclohexyl or cycloheptyl and R¹⁸ is hydrogen or hydroxy.

11. The compound of claim 1, or a pharmaceutically acceptable salt thereof, wherein —

CH(OH)CH₂CH₂NH₂.

12. The compound of claim 3, or a pharmaceutically acceptable salt thereof, wherein R⁵ is CH₂CR¹⁶R¹⁷R¹⁸;

R¹⁸ is hydrogen or hydroxyl and R¹⁶ and R¹⁷ are C_1-4 alkyl or are joined to form a carbocyclic ring of 5, 6, 7 or 8 ring carbon atoms optionally substituted by one or two moieties selected from halogen atoms, C_1-4 alkoxy groups, or C_1-4 alkyl groups.

13. The compound of claim 3, or a pharmaceutically acceptable salt thereof, wherein R¹⁷ and R¹⁸ are C_1-4 alkyl or are joined to form a carbocyclic ring of 5, 6, 7 or 8 ring carbon atoms.

14. The compound of claim 3, or a pharmaceutically acceptable salt thereof, wherein CR¹⁷R¹⁸ form a cyclohexyl.

15. The compound of claim 3, or a pharmaceutically acceptable salt thereof, wherein:

R¹⁴ is hydrogen;

R¹⁵ is hydrogen;

R¹⁸ is hydrogen or hydroxyl;

R¹⁶ and R¹⁷ are C_1-4 alkyl or are joined to form a carbocyclic ring of 5, 6, 7 or 8 carbon atoms optionally substituted by one or two moieties selected from halogen atoms or C_1-4 alkoxy groups;

R³ is hydrogen;

R⁴ is hydrogen;

R³³ is halogen, C_1-4 alkoxy, CO₂C_1-4 alkyl and n is 1.

17. A pharmaceutical composition comprising a pharmaceutically acceptable carrier and a compound as defined in claim 1 or 16, or a pharmaceutically acceptable salt thereof.
