Abstract

The present invention relates generally to compositions and methods for treating neurodegenerative diseases and disorders, particularly ophthalmic diseases and disorders. Provided herein are alkoxyl derivative compounds and pharmaceutical compositions comprising these compounds. The subject compositions are useful for treating and preventing ophthalmic diseases and disorders, including age-related macular degeneration (AMD) and Stargardt'"'"'s Disease.

Claims

1. A compound having a structure of Formula (IIa):

2. The compound of claim 1, or a pharmaceutically acceptable salt thereof, wherein each of R₃, R₄, R₁₄ and R₁₅ is hydrogen.

3. The compound of claim 1, or a pharmaceutically acceptable salt thereof, wherein

each of R₁, R₂, R₉ and R₁₀ is independently hydrogen, halogen, alkyl or —

OR₆, wherein R₆ is hydrogen or alkyl;

or R₉ and R₁₀ together form oxo;

R₁₆ and R₁₇ together with the carbon to which they are attached form a heterocyclyl having at least one oxygen ring atom, and R₁₈ is hydrogen, hydroxy or alkoxy.

4. The compound of claim 3, or a pharmaceutically acceptable salt thereof, wherein R₉ is —

OH, and R₁₀ is hydrogen.

5. The compound of claim 1, or a pharmaceutically acceptable salt thereof, wherein R₁₆ and R₁₇ together with the carbon to which they are attached form a heterocyclyl having at least one oxygen ring atom.

6. The compound of claim 5, or a pharmaceutically acceptable salt thereof, wherein each of R₃, R₄, R₁₄ and R₁₅ is hydrogen.

7. The compound of claim 5, or a pharmaceutically acceptable salt thereof, wherein,

each of R₁, R₂, R₉ and R₁₀ is independently hydrogen, halogen, alkyl or —

OR₆, wherein R₆ is hydrogen or alkyl;

or R₉ and R₁₀ together form oxo;

R₁₆ and R₁₇ together with the carbon to which they are attached form a heterocyclyl having at least one oxygen ring atom, and

R₁₈ is hydrogen, hydroxy or alkoxy.

8. The compound of claim 7, or a pharmaceutically acceptable salt thereof, wherein R₉ is —

OH, and R₁₀ is hydrogen.

9. The compound of claim 8, or a pharmaceutically acceptable salt thereof, wherein each of R₁₄ and R₁₅ is hydrogen.

10. The compound of claim 9, or a pharmaceutically acceptable salt thereof, wherein R₁₈ is hydrogen.

11. A pharmaceutical composition comprising a pharmaceutically acceptable carrier, and a compound of claim 1, or a tautomer, a mixture of tautomers, stereoisomer, pharmaceutically acceptable salt, or N-oxide thereof.
