Abstract

A method includes producing libraries of nanoparticles having highly controlled properties, which can be formed by mixing together two or more macromolecules in different ratios. One or more of the macromolecules may be a polymeric conjugate of a moiety to a biocompatible polymer. The nanoparticle may contain a drug. The moiety may include a polypeptide or a polynucleotide, such as an aptamer. The moiety may be a targeting moiety, an imaging moiety, a chelating moiety, a charged moiety, or a therapeutic moiety. Another aspect is directed to systems and methods of producing such polymeric conjugates. In some embodiments, a solution containing a polymer is contacted with a liquid, such as an immiscible liquid, to form nanoparticles containing the polymeric conjugate. Other methods use such libraries, use or administer such polymeric conjugates, or promote the use of such polymeric conjugates. Kits involving such polymeric conjugates are also described.

Claims

1. A method of making nanoparticles, comprising:

providing a solution comprising a first population of amphiphilic macromolecules comprising a first hydrophobic portion and a second hydrophilic portion and a second population of amphipilic macromolecules comprising a first hydrophobic portion, a second hydrophilic portion, and a third portion comprising a moiety selected from the group consisting of a targeting moiety, an imaging moiety, a chelating moiety, a moiety having multiple charge groups, and a diagnostic or therapeutic moiety,

wherein the first, second, or both portions of the amphiphilic macromolecules comprise a biodegradable polymer;

contacting the solution with a polymer nonsolvent under conditions wherein the macromolecules orient to form the nanoparticles having the hydrophobic portion on the inside of the particles and the hydrophilic portions and moieties on the surface of the nanoparticle.

2. The method of claim 1, wherein the moiety has a molecular weight of at least about 1000 Da.

3. The method of claim 1, wherein the moiety has a molecular weight of no more than about 1000 Da.

4. The method of claim 1, wherein at least a portion of the macromolecule is hydrolyzable.

5. The method of claim 1, wherein the targeting moiety comprises a molecule selected from the group consisting of a polypeptide, a nucleic acid, a fatty acid, a carbohydrate, a peptidoglycan, and a glycopeptide.

6. The method of claim 1, wherein the targeting moiety comprises an antibody or an antibody fragment.

7. The method of claim 1, wherein the targeting moiety is able to specifically bind to a molecule on the surface of a cell.

The method of claim 1, wherein the targeting moiety is able to specifically bind to a biological entity selected from the group consisting of bacterial membrane proteins, cytokines, chemokines, growth factors, insulin, erythropoietin, tumor necrosis factor, glycoproteins, adhesion molecules, fibronectins, laminin, and antigens.

9. The method of claim 1, wherein the solution further comprises an antigen.

10. The method of claim 1, wherein the nanoparticles have an average characteristic dimension of less than about 150 nm.

11. The method of claim 1, wherein the hydrophilic portion is a poly(alkylene glycol).

12. The method of claim 11, wherein the poly(alkylene glycol) comprises polyethylene glycol).

13. The method of claim 1, wherein the solution comprising the macromolecule is an organic solution and the polymer nonsolvent is an aqueous solution.

14. The method of claim 1, wherein the biodegradable polymer comprises poly(lactide-co-glycolide).

15. The method of claim 1, further comprising adding a therapeutic, prophylactic, or diagnostic agent prior to forming the nanoparticles so that the agent is encapsulated within the nanoparticles.

16. The method of claim 15, wherein the agent is a chemotherapeutic for the treatment of cancer.

17. The method of claim 1, further comprising adding an interfering RNA prior to forming the nanoparticles.

18. The method of claim 15, further comprising adding a peptide or a protein prior to forming the nanoparticles.

19. The method of claim 15, comprising adding a diagnostic agent with the therapeutic agent prior to forming the nanoparticles.

20. The method of claim 1, wherein the hydrophobic portion of the first population of amphiphilic macromolecules differs from the hydrophobic portion of the second population of amphiphilic macromolecules.

21. The method of claim 1, wherein the hydrophilic portion of the first population of amphiphilic macromolecules differs from the hydrophilic portion of the second population of amphiphilic macromolecules.

22. The method of claim 1, wherein the ratio of the hydrophobic portion to the hydrophilic portion of the first population of amphiphilic macromolecules differs from the ratio of the hydrophobic portion to the hydrophilic portion of the second population of amphiphilic macromolecules.
