Abstract

This invention comprises the novel compounds of formula (I)

wherein n, R¹, R², R³, R⁴, Q, X, Y, Z and

have defined meanings, having histone deacetylase inhibiting enzymatic activity; their preparation, compositions containing them and their use as a medicine.

Claims

1. A compound of formula (I),

N(OH)R⁷ wherein R⁵ and R⁶ are each independently selected from hydrogen, hydroxy, hydroxyC_1-6alkyl or aminoC_1-6alkyl;

R² is hydrogen, halo, hydroxy, amino, nitro, C_1-6alkyl, C_1-6alkyloxy, trifluoromethyl or di(C_1-6alkyl)amino;

R³ is C_1-6alkyl, arylC_2-6alkenediyl, furanylcarbonyl, naphthalenylcarbonyl, —

C(O)phenylR⁹, C_1-6alkylaminocarbonyl, aminosulfonyl, arylaminosulfonyl, aminosulfonylamino, di(C_1-6alkyl)aminosulfonylamino, di(C_1-6alkyl)aminoC_1-6alkyl, C_1-12alkylsulfonyl, di(C_1-6alkyl)aminosulfonyl or pyridinylcarbonyl wherein each R⁹ is independently selected from phenyl;

3. The compound as claimed in claim 1 wherein

R² is hydrogen or nitro;

R³ is C_1-6alkyl, arylC_2-6alkenediyl, furanylcarbonyl, naphthalenylcarbonyl, C_1-6alkylaminocarbonyl, aminosulfonyl, di(C_1-6alkyl)aminosulfonylaminoC_1-6alkyl, di(C_1-6alkyl)aminoC_1-6alkyl, C_1-12alkylsulfonyl, di(C_1-6alkyl)aminosulfonyl, trihaloC_1-6alkylsulfonyl, di(aryl)C_1-6alkylcarbonyl, thiophenylC_1-6alkylcarbonyl, pyridinylcarbonyl or arylC_1-6alkylcarbonyl; and

R⁴ is hydrogen.

4. The compound as claimed in claim 1 wherein

n is 1;

R¹ is —

C(O)NH(OH);

R² is hydrogen;

R³ is naphthalenylcarbonyl, C_1-12alkylsulfonyl or di(aryl)C_1-6alkylcarbonyl; and

R⁴ is hydrogen.

5. The compound according to claim 1 that is

6. A pharmaceutical composition comprising pharmaceutically acceptable carriers and as an active ingredient, a therapeutically effective amount of a compound as claimed in claim 1.

7. A process of preparing a pharmaceutical composition as claimed in claim 6 comprising intimately mixing the pharmaceutically acceptable carriers and the compound.

8. A method of treating ovarian carcinoma in a patient comprising administering to the patient a therapeutically effective amount of a compound of claim 1.

9. A process for preparing a compound as claimed in claim 1, comprising

reacting an intermediate of formula (II) with an acid, yielding a hydroxamic acid of formula (I-a)

10. The process of claim 9, wherein the acid is trifluoro acetic acid.

11. A method of detecting or identifying a histone deactylase (HDAC) in a biological sample comprising detecting or measuring the formation of a complex between a labelled compound as defined in claim 1 and a HDAC.

12. A combination of anti-cancer agents and a compound of claim 1.
