Abstract

The invention relates to compositions and methods for the preparation, manufacture and therapeutic use of polynucleotides, primary transcripts and mmRNA molecules.

Claims

1. An isolated polynucleotide which is an mRNA, said mRNA comprising:

a first region of linked nucleosides, said first region encoding SEQ ID NO;

2626 and said first region having at least 85% identity to SEQ ID NO;

15265.

2. A pharmaceutical composition comprising the isolated polynucleotide of claim 1 and a pharmaceutically acceptable excipient.

3. The pharmaceutical composition of claim 2, wherein the excipient is lipid nanoparticle.

4. The isolated polynucleotide of claim 1, wherein the mRNA comprises at least one untranslated region 5′

relative to the first region and at least one untranslated region 3′

relative to the first region.

5. The isolated polynucleotide of claim 4, wherein the 5′

untranslated region is heterologous to the first region of the mRNA.

6. The isolated polynucleotide of claim 4, wherein the 3′

untranslated region is heterologous to the first region of the mRNA.

7. The isolated polynucleotide of claim 4, wherein the 5′

untranslated region and the 3′

untranslated region are heterologous to the first region of the mRNA.

8. The isolated polynucleotide of claim 4, wherein the mRNA comprises at least two stop codons.

9. The isolated polynucleotide of claim 1, wherein the coding region is selected from the group consisting of SEQ ID NO:
