- (a) a bottom substrate and a removable top substrate;
(b) an array of droplet operations electrodes arranged on the bottom substrate;
wherein the top substrate is a self-contained replaceable/removable cartridge for a droplet actuator-based assay and an interface material comprising a dielectric layer disposed between the bottom substrate and the cartridge in such a way as form a reverse impression of the array of droplet operations electrodes on the bottom substrate.

15. A droplet actuator device for conducting droplet operations, comprising:

(a) a bottom substrate having an array of droplet operations electrodes arranged thereon; (b) a top substrate, wherein the top substrate is a self-contained replaceable/removable cartridge for a droplet actuator-based assay; and (c) an interface material comprising a dielectric layer disposed between the bottom substrate and the cartridge in such a way as to form a reverse impression of the array of droplet operations electrodes on the bottom substrate, wherein the interface material provides a tight coupling between the bottom substrate and the cartridge when the cartridge is assembled on the bottom substrate.
