Abstract

The present compositions and methods relate to a beta-mannanase from Bacillus hemicellulosilyticus, polynucleotides encoding the beta-mannanase, and methods of make and/or use thereof. Formulations containing the beta-mannanase are suitable for use in hydrolyzing lignocellulosic biomass substrates, especially those comprising a measurable level of galactoglucomannan (GGM) and/or glucomannan (GM).

Claims

1. A recombinant nucleic acid which comprises a nucleic acid encoding a recombinant polypeptide comprising the amino acid sequence of SEQ ID NO:

2 or SEQ ID NO;

3 and a heterologous nucleic acid, wherein the recombinant polypeptide has beta-mannanase activity, and wherein said nucleic acid encoding the recombinant polypeptide is operably linked to the heterologous nucleic acid.

2. The recombinant nucleic acid of claim 1, wherein the heterologous nucleic acid encodes a signal peptide sequence selected from any one of SEQ ID NOs:

13-41.

3. A host cell comprising the recombinant nucleic acid of claim 1, wherein the host cell is a microorganism.

4. The host cell of claim 3 wherein the heterologous nucleic acid encodes a signal peptide sequence.

5. The host cell of claim 4 wherein the signal peptide sequence is selected from any one of SEQ ID NOs:

13-41.

6. The host cell of claim 3 wherein the host cell is a bacterial cell or a fungal cell.

7. The host cell of claim 3 wherein the host cell is Trichoderma reesei.

8. The recombinant nucleic acid of claim 1, wherein the recombinant polypeptide comprises the amino acid sequence of SEQ ID NO:
