Abstract

The present invention relates to recombinant gelatins and compositions thereof, and methods of producing and using the same.

Claims

1. A recombinant gelatin comprising an amino acid sequence selected from the group consisting of SEQ ID NOs:

15, 16, 17, 19, 20, 21, 22, 23, 24, 25, 30, 31, and 33.

2. An isolated and purified polynucleotide encoding an amino acid sequence selected from the group consisting of SEQ ID NOs:

15, 16, 17, 18, 19, 20, 21, 22, 23, 24, 25, 30, 31, and 33.

3. An expression vector comprising the polynucleotide of claim 2.

4. A host cell comprising the polynucleotide of claim 2.

5. The host cell of claim 4, wherein the host cell is a prokaryotic cell.

6. The host cell of claim 4, wherein the host cell is a eukaryotic cell.

7. The eukaryotic cell of claim 6, wherein the eukaryotic cell is selected from the group consisting of a yeast cell, an animal cell, an insect cell, a plant cell, and a fungal cell.

8. A transgenic animal comprising the polynucleotide of claim 2.

9. A transgenic plant comprising the polynucleotide of claim 2.

10. A recombinant gelatin comprising an amino acid sequence selected from the group consisting of SEQ ID NOs:

26, 27, and 28.

11. A method of producing recombinant gelatin, the method comprising:

(a) providing recombinant collagen or procollagen or fragments or variants thereof, and

(b) processing recombinant collagen or procollagen or fragments or variants thereof to produce recombinant gelatin.

12. The method of claim 11, wherein the recombinant collagen is recombinant human collagen.

13. The method of claim 11, wherein the recombinant collagen or procollagen is produced by co-expressing at least one polynucleotide encoding a collagen or procollagen and at least one polynucleotide encoding a collagen post-translational enzyme or subunit thereof.

14. The method of claim 13, wherein the post-translational enzyme is prolyl hydroxylase.

15. A method of producing recombinant gelatin, the method comprising expressing in a host cell a polynucleotide sequence that encodes at least one collagenous domain and that does not encode naturally occurring collagen.

16. The method of claim 15, wherein the recombinant gelatin is producing by co-expressing in a host cell the polynucleotide sequence that encodes at least one collagenous domain and that does not encode naturally occurring collagen and at least one polynucleotide sequence that encodes a post-translational enzyme or subunit thereof.

17. The method of claim 16, wherein the post-translational enzyme is prolyl hydroxylase.
