Abstract

A biological soil detector is provided, comprising: a solid support member; a specific indicator immobilized on the solid support member; and a retaining member holding the solid support thereon, the retaining member and solid support configured to facilitate contact between the solid support a surface. A method for the use of the foregoing detector is also provided, the method comprising: contacting the foregoing biological soil detector with a surface wherein the contacting initiates the modification of the specific indicator in the presence of bio soil; withdrawing the biological soil detector from the surface; and inspecting the solid support member of the detector for a detectable response to thereby determine whether the specific indicator immobilized on the solid support has interacted with biological soil. In still another aspect, the invention provides biological soil detector and a method for using the detector wherein the detector comprises: a solid support member treated with at least one amine or quaternary functional coating; and a specific indicator immobilized on the coating.

Claims

1. A biological soil detector, comprising:

A solid support member;

A specific indicator immobilized on the solid support member; and

A retaining member holding the solid support thereon, the retaining member and solid support configured to facilitate contact between the solid support a surface.

2. The biological soil detector of claim 1 wherein the solid support comprises a first material selected from the group consisting of at least one polymer;

inorganic material; and

mixed organic and inorganic material.

The biological soil detector of claim 2 wherein the polymer is selected from polymers containing functional groups, the functional groups selected from the group consisting of carboxyl and salts thereof, aldehydes, sulfonic acid and salts thereof, phosphonic acid and salts thereof, alcohol, primary amine, secondary amine, tertiary amine, amide, imide, quaternized ammonium, sulfonium, phosphonium, pyridine, cyclic amido, oxyalkylene, imidazoles and combinations of two or more of the foregoing.

4. The biological soil detector of claim 3 wherein the polymers containing functional groups comprise polymers and copolymers selected from the group consisting of carboxyl containing polymers;

polyalkoxylates;

poly(meth)acrylates;

polyvinyl alcohol;

polyethylene-vinyl alcohol copolymer;

polyurethane;

polyurea;

polyester;

polyamide;

polyimide;

polyether;

cellulose;

rayon;

polyphosphate;

polypeptide;

polyacrylonitrile;

polyacrylamide;

polycarbonate;

polyethersulfone; and

combinations of two or more of the foregoing.

5. The biological soil detector of claim 2 wherein the inorganic material is selected from the group consisting of metal oxide, hydrate, metal-hydroxyl and combinations of two or more of the foregoing.

6. The biological soil detector of claim 2 wherein the mixed organic and inorganic material is selected from the group consisting of polymeric composites, ceramers, and combinations thereof.

7. The biological soil detector of claim 1 wherein the solid support member comprises a first material selected from the group consisting of at least one polymer, inorganic material, and mixed organic and inorganic material;

and a second material attached to the first material.

The biological soil detector of claim 7 wherein the first material is selected from polypropylene, polyethylene, polyvinylidene fluoride, tetrafluoroethylene hexafluoropropylene vinylidene fluoride, polyurethane, polyurea, polyester, polyvinyl acetate, polyamide, polyimide, poly(meth)acrylate, polyethersulfone, glass, silica, cellulosics, rayon, polycarbonate, polyvinyl alcohol, polystyrene, and combinations of the foregoing.

9. The biological soil detector of claim 8 wherein the second material is hydrophilic and is selected from the group consisting of monomer, polymer, reactive metal alkoxide and combinations of two or more of the foregoing.

The biological soil detector of claim 9 wherein the second material comprises one or more functional groups selected from carboxyl and salts thereof, aldehyde, sulfonic acid and salts thereof, phosphonic acid and salts thereof, alcohol, primary amine, secondary amine, tertiary amine, amide, imide, quaternized ammonium, sulfonium, phosphonium, pyridine, cyclic amido, oxyalkylene, co-saccharinamidoundecylsiloxane, glycidyl, succinimido, imidazoles and combinations of two or more of the foregoing.

The biological soil detector of claim 2 wherein the solid support member comprises a material selected from the group consisting of films, nonwoven material, woven material, knitted material, reticulated foam, open-celled foam, porous ceramic inorganic frit, fiber, particle-coated supports, sintered particles, sintered fiber, sponge, and polymer membrane.

The biological soil detector of claim 11 wherein the polymer membrane comprises a thermally induced phase separation membrane comprising a material selected from the group consisting of high density polyethylene, polypropylene, polyvinylidenefluoride, polyethylene-vinyl copolymer and combinations of two or more of the foregoing.

13. The biological soil detector of claim 2 wherein the solid support member further comprises a hydrophilic coating.

14. The biological soil detector of claim 1 wherein the specific indicator is selected from the group consisting of 5-bromo-4-chloro-3-indolyl β

-D-glucopyranoside;

5-bromo-4-chloro-3-indolyl β

-D-galactopyranoside;

5-bromo-4-chloro-3-indolyl phosphate;

5-bromo-6-chloro-3-indolyl-β

-D-glucopyranoside;

5-bromo-6-chloro-3-indolyl-β

-D-galactopyranoside;

5-bromo-6-chloro-3-indolyl phosphate;

4-methylumbelliferyl-β

-D-glucopyranoside;

4-methylumbelliferyl-β

-D-galactopyranoside;

4-methylumbelliferyl-phosphate, esculin, orthophthaldialdehyde (OPA), polydiacetylenes;

Bradford assay;

Lowry, Biuret assay;

bicinchoninic acid (BCA); and

combinations of two or more of the foregoing

15. The biological soil detector of claim 1 wherein the specific indicator comprises NBT in combination with on or more indolyl functional indicators selected from the group consisting of 5-bromo-4-chloro-3-indolyl β

-D-glucopyranoside;

5-bromo-4-chloro-3-indolyl β

-D-galactopyranoside;

5-bromo-4-chloro-3-indolyl phosphate;

5-bromo-6-chloro-3-indolyl-p-D-glucopyranoside;

5-bromo-6-chloro-3-indolyl-β

-D-galactopyranoside;

5-bromo-6-chloro-3-indolyl phosphate.

16. The biological soil indicator of claim 15, wherein the specific indicator further comprises monovalent or divalent metal ions selected from sodium, potassium, zinc, manganese, magnesium, calcium and combinations of one or more of the foregoing.

17. The biological soil indicator of claim 1 wherein the specific indicator is selected to detect a component of biological soil selected from the group consisting of protein, endotoxin, enzyme, nucleotide and combinations of two or more of the foregoing.

18. The biological soil indicator of claim 17 wherein the nucleotide is adenosine triphosphate.

19. The biological soil indicator of claim 17 wherein the enzyme comprises galactosidases, phosphatases, glucosidases, and lactosidases and combinations thereof.

20. The biological soil indicator of claim 1 wherein specific indicator is selected to detect sulfatases or fatty acid esterases.

21. The biological soil detector of claim 1 wherein the retaining member is dimensioned to fit within a channel, the retaining member comprising a first end and a second end and an elongate body portion extending between the first end and the second end.

22. The biological soil detector of claim 1 wherein the solid support member is releasably affixed to the retaining member.

23. A method for detecting biological soil associated with a surface, the method comprising:

Contacting the biological soil detector of claim 1 with a surface wherein the contacting initiates the modification of the specific indicator in the presence of bio soil;

Withdrawing the biological soil detector from the surface;

Inspecting the solid support member of the detector for a detectable response to thereby determine whether the specific indicator immobilized on the solid support has interacted with biological soil.

24. The method of claim 23 wherein the specific indicator is selected to detect a component of biological soil selected from the group consisting of protein, endotoxin, enzyme, nucleotide and combinations of two or more of the foregoing.

25. The method of claim 24 wherein the nucleotide is adenosine triphosphate.

26. The method of claim 24 wherein the enzyme comprises galactosidase, phosphatase, glucosidase, and lactosidase.

27. The method of claim 23 wherein the specific indicator is selected to detect sulfatases or fatty acid esterases.

28. A method according to claim 23 wherein the surface is a channel of a medical device.

29. A method according to claim 28 wherein the medical device is an endoscope.

30. A method according to claim 23 wherein the specific indicator is selected from the group consisting of 5-bromo-4-chloro-3-indolyl β

-D-glucopyranoside;

5-bromo-4-chloro-3-indolyl β

-D-galactopyranoside;

5-bromo-4-chloro-3-indolyl phosphate;

5-bromo-6-chloro-3-indolyl-β

-D-glucopyranoside;

5-bromo-6-chloro-3-indolyl-β

5-bromo-4-chloro-3-indolyl phosphate;

32. The method of claim 31 wherein the specific indicator further comprises monovalent or divalent metal ions selected from sodium, potassium, zinc, manganese, magnesium, calcium and combinations of one or more of the foregoing.

33. A method according to claim 23 wherein inspecting the solid support member for a detectable response comprises visually examining the solid support for a change in color.

34. A method according to claim 23 wherein inspecting the solid support member for a detectable response comprises examining the solid support member for a change in fluorescence.

35. A method according to claim 34 wherein a detectable response is present within 10 minutes following the contacting of the biological soil detector with a surface.

36. A method according to claim 34 wherein a detectable response is present within 5 minutes following the contacting of the biological soil detector with a surface.

37. A method according to claim 34 wherein a detectable response is present in less than one minute following the contacting of the biological soil detector with a surface.

38. A biological soil detector, comprising:

A solid support member treated with at least one amine or quaternary functional coating; and

A specific indicator immobilized on the coating.

39. The biological soil detector of claim 38 wherein the solid support member is a nonwoven material.

40. The biological soil detector of claim 39 wherein the nonwoven material comprises a cellulosic material.

41. The biological soil detector of claim 39 wherein the nonwoven material comprises 70% rayon and 30% polyester.

42. The biological soil detector of claim 38 wherein the solid support member comprises a material selected from the group consisting of films, nonwoven material, woven material, knitted material, reticulated foam, open-celled foam, fiber, particle-coated supports, sponge, and polymer membrane.

43. The biological soil detector of claim 42 wherein the polymer membrane comprises a thermally induced phase separation membrane comprising a material selected from the group consisting of high density polyethylene, polypropylene, polyvinylidenefluoride, polyethylene-vinyl copolymer and combinations of two or more of the foregoing.

44. The biological soil detector of claim 38 wherein the solid support member further comprises a hydrophilic coating.

45. The biological soil detector of claim 38 wherein the specific indicator is selected from the group consisting of 5-bromo-4-chloro-3-indolyl β

-D-glucopyranoside;

5-bromo-4-chloro-3-indolyl β

-D-galactopyranoside;

5-bromo-4-chloro-3-indolyl phosphate;

5-bromo-6-chloro-3-indolyl-β

-D-glucopyranoside;

5-bromo-6-chloro-3-indolyl-β

-D-galactopyranoside;

5-bromo-6-chloro-3-indolyl phosphate;

4-methylumbelliferyl-β

46. The biological soil detector of claim 38 wherein the specific indicator comprises NBT in combination with one or more indolyl functional indicators selected from the group consisting of 5-bromo-4-chloro-3-indolyl β

-D-glucopyranoside;

5-bromo-4-chloro-3-indolyl β

-D-galactopyranoside;

5-bromo-4-chloro-3-indolyl phosphate;

5-bromo-6-chloro-3-indolyl-β

-D-glucopyranoside;

5-bromo-6-chloro-3-indolyl-β

-D-galactopyranoside;

5-bromo-6-chloro-3-indolyl phosphate.

47. The biological soil indicator of claim 46, wherein the specific indicator further comprises monovalent or divalent metal ions selected from sodium, potassium, zinc, manganese, magnesium, calcium and combinations of one or more of the foregoing.

48. The biological soil indicator of claim 38 wherein the specific indicator is selected to detect a component of biological soil selected from the group consisting of protein, endotoxin, enzyme, nucleotide and combinations of two or more of the foregoing.

49. The biological soil indicator of claim 48 wherein the nucleotide is adenosine triphosphate.

50. The biological soil indicator of claim 48 wherein the enzyme comprises galactosidases, phosphatases, glucosidases, and lactosidases and combinations thereof.

51. The biological soil indicator of claim 38 wherein specific indicator is selected to detect sulfatases or fatty acid esterases.

52. The biological soil detector of claim 38 wherein the solid support member further comprises an amine-containing silane.

53. The biological soil detector of claim 52 wherein the amine-containing silane is selected from the group consisting of (N-trimethoxysilylpropyl) polyethyleneimine, 3-aminopropyltriethoxysilane, N-trimethoxysilylpropyl-N,N,N-trimethylammonium chloride and combinations of two or more of the foregoing.

54. A method for detecting biological soil associated with a surface, the method comprising:

Contacting the biological soil detector of claim 38 with a surface wherein the contacting initiates the modification of the specific indicator in the presence of bio soil;

Withdrawing the biological soil detector from the surface;

Inspecting the solid support member of the detector for a detectable response to thereby determine whether the specific indicator immobilized on the solid support has interacted with biological soil.

55. The method of claim 54 wherein the specific indicator is selected to detect a component of biological soil selected from the group consisting of protein, endotoxin, enzyme, nucleotide and combinations of two or more of the foregoing.

56. The method of claim 55 wherein the nucleotide is adenosine triphosphate.

57. The method of claim 55 wherein the enzyme comprises galactosidase, phosphatase, glucosidase, and lactosidase.

58. The method of claim 54 wherein the specific indicator is selected to detect sulfatases or fatty acid esterases.

59. A method according to claim 54 wherein the specific indicator is selected from the group consisting of 5-bromo-4-chloro-3-indolyl β

-D-glucopyranoside;

5-bromo-4-chloro-3-indolyl β

-D-galactopyranoside;

5-bromo-4-chloro-3-indolyl phosphate;

5-bromo-6-chloro-3-indolyl-β

-D-glucopyranoside;

61. A method according to claim 54 wherein inspecting the solid support member for a detectable response comprises visually examining the solid support for a change in color.

62. A method according to claim 61 wherein a detectable response is present within 10 minutes following the contacting of the biological soil detector with a surface.

63. A method according to claim 61 wherein a detectable response is present within 5 minutes following the contacting of the biological soil detector with a surface.

64. A method according to claim 61 wherein a detectable response is present in less than one minute following the contacting of the biological soil detector with a surface.
