Abstract

A method of non-invasively measuring the presence, absence, or concentration of one or more analytes in a tissue of a subject includes exposing at least a portion of a tissue of the subject to electromagnetic radiation; obtaining a measurement of electromagnetic radiation from the tissue; and determining the presence, absence, or concentration of one or more analytes by determining a radiation signature of the electromagnetic radiation; correlating the radiation signature of the electromagnetic radiation with at least one of concentration, presence, and absence of one or more analytes by utilizing a mathematic transformation such as a transformation based on a 1^st or higher derivative of the measurement to determine the presence, absence, or concentration of one or more analytes in the subject.

Claims

1. A method of non-invasively measuring the presence, absence, or concentration of one or more analytes in a tissue of a subject, comprising:

exposing at least a portion of a tissue of the subject to electromagnetic radiation;

obtaining a measurement of electromagnetic radiation from the tissue; and

determining the presence, absence, or concentration of one or more analytes by

determining a radiation signature of the electromagnetic radiation;

correlating the radiation signature of the electromagnetic radiation with the presence, absence, or concentration of one or more analytes by utilizing a function based on a mathematical transformation of the measurement to determine the presence, absence, or concentration of one or more analytes in the subject.

The method of claim 1, wherein the one or more analytes include a member from the group consisting of metabolic compounds or substances, neurotransmitters, carbohydrates, sugars, glucose, proteins, glycated proteins, peptides, amino acids, fats, fatty acids, triglycerides, polysaccharides, alcohols, ethanol, toxins, hormones, nucleic acids, vitamins, bacteria-related substances, fungus-related substances, virus-related substances, parasite-related substances, pharmaceutical compounds, non-pharmaceutical compounds, pro-drugs, drugs, and any precursor, metabolite, degradation product or surrogate marker.

3. The method of claim 2, wherein the one or more analytes include glucose.

4. The method of claim 1, wherein the non-invasive method does not contact the subject or tissue.

5. The method of claim 1, wherein the electromagnetic radiation is infrared radiation.

6. The method of claim 1, wherein the tissue is an ocular element.

7. The method of claim 1, wherein the ocular element is at least one of a conjunctiva and a tear layer.

8. The method of claim 1, wherein a 1st or higher derivative of reflectance measurements is used to select or measure one or more ideal wavelengths of electromagnetic radiation from the tissue.

9. The method of claim 7, wherein detecting electromagnetic radiation further comprises filtering the electromagnetic radiation from the tissue to select or measure the one or more ideal wavelengths of the electromagnetic radiation from the tissue.

10. The method of claim 1, wherein the determining step further comprises using a microprocessor.

11. The method of claim 1, wherein the measurement is a specular reflection measurement, and detecting includes detecting electromagnetic radiation with specular reflection optics.

12. The method of claim 1, wherein the measurement is a diffuse reflection measurement, and detecting includes detecting electromagnetic radiation with diffuse reflection optics.

13. An instrument for non-invasively measuring the presence, absence, or concentration of one or more analytes in a tissue of a subject, comprising:

a. a radiation source configured to expose at least a portion of the tissue of the subject to electromagnetic radiation;

b. a radiation detector configured to obtain a measurement of electromagnetic radiation from the tissue; and

c. a microprocessor for determining the presence, absence, or concentration of one or more analytes by determining a radiation signature of the electromagnetic radiation, and correlating the radiation signature of the electromagnetic radiation with the presence, absence, or concentration of one or more analytes by utilizing a mathematical transformation of the measurement to determine the presence, absence, or concentration of one or more analytes in the subject.

The instrument of claim 13, wherein the one or more analytes include a member from the group consisting of metabolic compounds or substances, neurotransmitters, carbohydrates, sugars, glucose, proteins, glycated proteins, peptides, amino acids, fats, fatty acids, triglycerides, polysaccharides, alcohols, ethanol, toxins, hormones, nucleic acids, vitamins, bacteria-related substances, fungus-related substances, virus-related substances, parasite-related substances, pharmaceutical compounds, non-pharmaceutical compounds, pro-drugs, drugs, and any precursor, metabolite, degradation product or surrogate marker.

15. The instrument of claim 14, wherein the one or more analytes include glucose.

16. The instrument of claim 13, wherein the instrument does not contact the subject or tissue.

17. The instrument of claim 13, wherein the electromagnetic radiation is infrared radiation.

18. The instrument of claim 13, wherein the tissue is an ocular element.

19. The instrument of claim 13, wherein the ocular element is at least one of a conjunctiva and a tear layer.

20. The instrument of claim 13, wherein the microprocessor utilizes a 1st or higher derivative of the measurement to select or measure one or more ideal wavelengths of the electromagnetic radiation from the tissue.

21. The instrument of claim 20, further including a filter to filter the reflected electromagnetic radiation from the tissue to select or measure the one or more ideal wavelengths of the electromagnetic radiation from the tissue, and the filter is at least one of a digital filter, an electronic filter, and an optical filter.

22. The instrument of claim 13, wherein the measurement is a specular reflection measurement, and the electromagnetic radiation wavelength detector includes specular reflection optics to take the specular reflection measurement.

23. The instrument of claim 13, wherein the measurement is a diffuse reflection measurement, and the electromagnetic radiation wavelength detector includes diffuse reflection optics to take the diffuse reflection measurement.

24. The instrument of claim 13, wherein the radiation source, the radiation detector, and the microprocessor are at least one of integrated together to form a single device and separated to form two or more separate devices.

25. A method of non-invasively measuring the presence, absence, or concentration of one or more analytes in a tissue of a subject, comprising:

using a non-invasive instrument to obtain a measurement of the presence, absence, or concentration of one or more analytes in the tissue of the subject without contacting the tissue;

determining the presence, absence, or concentration of one or more analytes in the tissue of the subject by utilizing a mathematical transformation based on a 1st or higher derivative of the measurement to determine the presence, absence, or concentration of one or more analytes in the tissue of the subject.

26. A method of non-invasively measuring the presence, absence, or concentration of one or more analytes in a tissue of a subject, comprising:

exposing at least a portion of the tissue of the subject to electromagnetic radiation without contact with the tissue;

obtaining a measurement of infrared radiation from the tissue without contact with the tissue; and

, determining the presence, absence, or concentration of one or more analytes in the tissue of the subject by

determining a radiation signature of the infrared radiation from the tissue;

correlating the radiation signature of the infrared radiation from the tissue with the presence, absence, or concentration of one or more analytes by utilizing a mathematical transformation based on a 1^st or higher derivative of the measurement to determine the presence, absence, or concentration of one or more analytes in the tissue of the subject.

27. The method of claim 26, wherein the electromagnetic radiation is mid-infrared radiation.

28. The method of claim 27, wherein the subject includes an eye with an ocular surface and ocular elements, and exposing includes exposing at least a portion of an ocular element of the subject to mid-infrared radiation without contact with the ocular surface.

29. An instrument for non-invasively measuring the presence, absence, or concentration of one or more analytes in a tissue of a subject, comprising:

a. a radiation source configured to expose at least a portion of the tissue of the subject to electromagnetic radiation;

b. a radiation detector configured to obtain a measurement of electromagnetic radiation from the tissue; and

c. hardware for determining the presence, absence, or concentration of one or more analytes by determining a radiation signature of the electromagnetic radiation, and correlating the radiation signature of the electromagnetic radiation with the presence, absence, or concentration of one or more analytes by utilizing analog different measurements to determine the presence, absence, or concentration of one or more analytes in the subject.

30. A method of correcting magnitude or focus variation in the non-invasive measurement of the presence, absence, or concentration of one or more analytes in a tissue of a subject, comprising:

using a non-invasive instrument to obtain a measurement of the presence, absence, or concentration of one or more analytes in the tissue of the subject;

correcting magnitude or focus variation in the measurement by utilizing a mathematical transformation based on a 1^st or higher derivative of the measurement;

determining the presence, absence, or concentration of one or more analytes in the tissue of the subject by

determining a signature of the corrected measurement of the presence, absence, or concentration of one or more analytes in the tissue;

correlating the signature of the corrected measurement with known signature information representative of the presence, absence, or concentration of one or more analytes to determine the presence, absence, or concentration of one or more analytes in the tissue of the subject.
