Abstract

Homogeneous preparations of IL-28A, IL-28B, and IL-29 have been produced by mutating one or more of the cysteine residues in the polynucleotide sequences encoding the mature proteins. The cysteine mutant proteins can be shown to either bind to their cognate receptor or exhibit biological activity. One type of biological activity that is shown is an antiviral activity.

Claims

1. An isolated polynucleotide encoding a polypeptide wherein the encoded polypeptide comprises amino acid residues 1-176 of SEQ ID NO:

2. The polynucleotide of claim 1 wherein the encoded polypeptide has anti-hepatitis activity.

3. The polynucleotide of claim 2 wherein the encoded polypeptide has anti-hepatitis B activity.

4. The polynucleotide of claim 2 wherein the encoded polypeptide has anti-hepatitis C activity.

5. The polynucleotide of claim 1 wherein the encoded polypeptide is covalently attached to a polyethylene glycol.

6. The polynucleotide of claim 5 wherein the polyethylene glycol is N-terminally attached.

7. The polynucleotide of claim 5 wherein the polyethylene glycol is monomethoxy-PEG propionaldehyde.

8. The polynucleotide of claim 5 wherein the polyethylene glycol has a molecular weight of about 20 kD or about 30 kD.

10. An expression vector comprising the following operably linked elements:

a transcription promoter;

a DNA segment encoding a polypeptide of claim 1;

and

a transcription terminator.

11. A cultured cell into which has been introduced an expression vector of claim 10, wherein the cell expresses the polypeptide encoded by the DNA segment.

12. A method of producing a polypeptide comprising:

culturing a cell into which has been introduced an expression vector of claim 10, wherein the cell expresses the polypeptide encoded by the DNA segment; and

recovering the expressed polypeptide.
