Abstract

A method of manufacturing a tissue matrix for implantation into a patient is disclosed. The method sets forth collecting embryonic stem cells from a placenta which has been treated to remove residual cord blood and seeding the collected stem cells onto or into a tissue matrix. The seeded tissue matrix is then implanted on or into a patient. The seeded tissue matrix made by the method of the present invention is also disclosed.

A method of making a tissue matrix, comprising seeding human placental stem cells into or onto a decellularized or synthetic tissue matrix, wherein said stem cells are from a human placenta that has been drained of cord blood and perfused to remove residual blood, and comprise stem cells that are CD34⁻

or OCT-4⁺.

25. The method of claim 24, wherein said placental stem cells are additionally CD10⁺, CD29⁺, CD44⁺, CD45⁻

, CD54⁺, CD90⁺, SH2⁺, SH3⁺, SH4⁺, SSEA3⁺ or SSEA4⁺, or any combination of the foregoing.

26. The method of claim 24, additionally comprising seeding CD34⁺ human placental stem cells onto said tissue matrix.

27. The method of claim 24, wherein said stem cells are obtained from a placenta that has been perfused for at least 2 hours.

28. The method of claim 24, wherein said stem cells are obtained from a placenta that has been perfused for at least 12 hours.

29. The method of claim 24, wherein said stem cells are obtained from a placenta that has been perfused for at least 24 hours.

30. The method of claim 24, wherein said decellularized tissue is contacted with serum and fibronectin prior to said seeding.

31. A decellularized or synthetic tissue matrix comprising human placental stem cells that are CD34⁻

or OCT-4⁺.

32. The tissue matrix of claim 31, wherein said placental stem cells are additionally CD10⁺, CD29⁺, CD44⁺, CD45⁻

, CD54⁺, CD90⁺, SH2⁺, SH3⁺, SH4⁺, SSEA3⁺ or SSEA4⁺, or any combination of the foregoing.

33. The tissue matrix of claim 31 additionally comprising CD34⁺ stem cells.

34. The tissue matrix of claim 31, wherein said tissue matrix comprises a synthetic tissue matrix.

35. The tissue matrix of claim 34, wherein said synthetic tissue matrix comprises polylactide-co-glycolide, a hydrated collagen lattice, or a collagen cross-linked with glycosaminoglycan.

36. The tissue matrix of claim 33, wherein said tissue matrix comprises a synthetic tissue matrix.

38. The tissue matrix of claim 31, wherein the tissue matrix comprises a decellularized tissue.

41. The tissue matrix of claim 33, wherein the tissue matrix comprises a decellularized tissue.

42. The tissue matrix of claim 41, wherein said decellularized tissue is non-human.

43. The tissue matrix of claim 41, wherein said tissue matrix comprises exogenous fibronectin, glycosaminoglycan, human serum, fetal bovine serum, fibroblast growth factor (FGF), platelet-derived growth factor (PDGF), transforming growth factors (TGF-β

), fibrillar collagen, or collagen fragments.

39. The tissue matrix of claim 39, wherein said decellularized tissue is non-human.

37. The tissue matrix of claim 39, wherein said synthetic tissue matrix comprises polylactide-co-glycolide, a hydrated collagen lattice, or a collagen cross-linked with glycosaminoglycan.

40. The tissue matrix of claim 39, wherein said tissue matrix comprises exogenous fibronectin, glycosaminoglycan, human serum, fetal bovine serum, fibroblast growth factor (FGF), platelet-derived growth factor (PDGF), transforming growth factors (TGF-β

), fibrillar collagen, or collagen fragments.
