Abstract

Reactive surfaces, substrates and methods of producing and using such substrates and surfaces are provided. The substrates and surfaces provide low density reactive groups preferably on an otherwise non-reactive surface for use in different applications including single molecule analyses.

36. A method of configuring a surface to provide a desired density of reactive moieties thereon, comprising treating the surface with a composition that substantially uniformly couples to the surface, the composition comprising a first component that doe'"'"'s not contain the reactive moiety and a second component that comprises the reactive moiety, the second component being present in the composition at a concentration relative to the first component so as to provide the reactive moieties on the surface at the desired density

37. The method of claim 36, wherein the surface comprises an observation region on a substrate.

38. The method of claim 36, wherein a catalytic moiety is coupled to the reactive moiety on the surface.

39. The method of claim 38, wherein the surface comprises an observation region on a substrate, and the desired density provides at least a 95% probability that there will be no more than one catalytic, moiety within the observation region.

40. The method of claim 39, wherein the observation region comprises an optically isolated region.

41. The method of claim 40, wherein the observation region comprises a bottom surface of a zero mode waveguide.

42. The method of claim 38, wherein the catalytic moiety comprises a nucleic acid polymerase.

43. The method of claim 42, wherein the nucleic acid polymerase is indirectly coupled to the second component through an intermediate linker component.

44. The method of claim 36, wherein the desired density is between about 1 reactive moiety per 50,000 nm² and 1 reactive moiety per 100 nm².

45. The method of claim 44, wherein the desired density is less than less than 1 reactive moiety per 1000 nm².

46. The method of claim 44, wherein the desired density is less than reactive moiety per 10,000 nm².

47. The method of claim 44, wherein the desired density is between about 1 reactive moiety per 2500 nm² and about 1 reactive moiety per 300 nm².
