Abstract

The present the invention provides methods, devices and systems for rapidly measuring analytes within a biological sample.

22. A test strip comprising:

a working electrode;

a counter electrode; and

a redox reagent system, the redox reagent system comprising a first redox couple and at least one secondary redox couple, wherein the first redox couple is selected and configured to mediate a target analyte in a sample and provide a primary peak and the at least one secondary redox couple selected and configured so as to provide a secondary peak.

23. The test strip of claim 22, wherein the test strip further comprises a reference electrode.

24. The test strip of claim 22, wherein the target analyte is glucose.

25. The test strip of claim 22, wherein the test strip is configured to determine concentration of the target analyte from 1 μ

L or less of sample.

26. The test strip of claim 25, wherein the test strip is configured to determine concentration of the target analyte from 0.5 μ

L or less of sample.

27. The test strip of claim 22, wherein the at least one secondary redox couple is configured to provide a secondary peak in response to a hematocrit level of the sample.

28. The test strip of claim 27, wherein the first redox couple and the at least one secondary redox couple are configured to provide similar response currents to the hematocrit level of the sample.

29. The test strip of claim 22, wherein the at least one secondary redox couple is configured to provide a secondary peak in response to the temperature of the sample.

30. The test strip of claim 29, wherein the first redox couple and the at least one secondary redox couple are configured to provide similar response currents to the temperature of the sample.

31. A method of measuring concentration of a target analyte in a sample, the method comprising:

contacting a sample of no more than 1 μ

L comprising the target analyte with a test strip comprising a working electrode;

a counter electrode; and

a redox reagent system, the redox reagent system comprising a first redox couple and at least one secondary redox couple, wherein the first redox couple is selected and configured to mediate a target analyte in a sample and provide a primary peak and the at least one secondary redox couple selected and configured so as to provide a secondary peak; and

determining the concentration of the target analyte from the no more than 1 μ

L of sample.

32. The method of claim 31, wherein the method further comprises:

applying a square wave excitation voltage to the test strip; and

measuring a response current, wherein the response current comprises a primary peak from mediation of the target analyte by the first redox couple and one or more secondary peaks from the at least one secondary redox couple.

33. The method of claim 32, wherein the one or more secondary peaks comprise a response current in response to a hematocrit level of the sample.

34. The method of claim 33, wherein the one or more secondary peaks comprise a response current in response to the temperature of the sample.

35. The method of claim 32, wherein measuring the response current further comprises:

decoupling the first peak from the one or more secondary peaks; and

correcting the first peak using the one or more secondary peaks.

36. The method of claim 31, wherein the target analyte is glucose.

37. The method of claim 31, wherein the method comprises contacting a sample of no more than 0.5 μ

L comprising the target analyte with the test strip.

38. The method of claim 31, wherein determining the concentration of the target analyte is performed within less than 1 second.

39. A method of measuring the concentration of glucose in a sample, the method comprising:

contacting a sample of no more than 1 μ

L comprising glucose with a test strip comprising a working electrode;

a counter electrode; and

a redox reagent system, the redox reagent system comprising a first redox couple and at least one secondary redox couple, wherein the first redox couple is selected and configured to mediate glucose in the sample and provide a primary peak and the at least one secondary redox couple selected and configured so as to provide a secondary peak;

applying a square wave excitation voltage to the test strip;

measuring a response current, wherein the response current comprises a primary peak from mediation of glucose by the first redox couple and one or more secondary peaks from the at least one secondary redox couple; and

determining the concentration of glucose from the no more than 1 μ

L of sample within less than 1 second.
