Abstract

Therapeutic stem cells and methods for their use and manufacture. Stem cells are produced under conditions in which the stem cells are exposed to at least one environmental factor, including decreased oxygen tension. The environmental factors and culture conditions of the invention produce stem cells having an enhanced therapeutic ability and enhanced proliferation in culture. Stem cells of the invention retain their plasticity through a higher number of cell passages relative to know methods of stem cell culture.

Claims

1. A kit for regenerative stem cell therapy comprising:

a. a therapeutic amount of neural stem cells in a pharmaceutically acceptable carrier; and

b. a therapeutic amount of mesenchymal stem cells in a pharmaceutically acceptable carrier;

c. wherein the neural stem cells and the mesenchymal stem cells have been exposed to at least one environmental factor.

2. The kit of claim 1, wherein said at least one environmental factor comprises reduced oxygen tension.

3. The kit of claim 1, wherein said at least one environmental factor comprises culturing the mesenchymal stem cells and the neural stem cells under reduced oxygen tension.

4. The kit of claim 3, wherein the neural stem cells and the mesenchymal stem cells are cultured in a culture medium comprising serum.

5. The kit of claim 4, wherein said culture medium comprises between about 0.1 and 2.0 percent serum.

6. A method for treating a medical disorder in a patient comprising:

a. administering to the patient a therapeutic amount of neural stem cells; and

b. administering to the patient a therapeutically effective amount mesenchymal stem cells;

c. wherein the neural stem cells and the mesenchymal stem cells have been contacted with at least one environmental factor.

7. The method of claim 6, wherein the at least one environmental factor comprises reduced oxygen tension.

8. The method of claim 6, wherein the at least one environmental factor comprises culturing the neural stem cells and the mesenchymal stem cells under reduced oxygen tension.

9. The method of claim 8, wherein the neural stem cells and the mesenchymal stem cells are cultured in a culture medium comprising serum.

10. The method of claim 9, wherein the medical disorder is selected from the group consisting of ischemic stroke, Alzheimer'"'"'s disease, spinal cord injury, Parkinson'"'"'s disease, traumatic brain injury, solid tumors, multiple sclerosis, amyotrophic lateral sclerosis, Huntington'"'"'s disease, muscular dystrophy, retinitis pigmentosa, cerebral palsy, dementia, neuropathies due to chemotherapy or radiation therapy, and neuromyopathy.

11. A method for treating a medical disorder in a patient comprising:

a. administering to the patient a therapeutically effective amount mesenchymal stem cells;

b. wherein the mesenchymal stem cells have been contacted with at least one environmental factor.

12. The method of claim 11, wherein the at least one environmental factor comprises reduced oxygen tension.

13. The method of claim 11, wherein the at least one environmental factor comprises culturing the neural stem cells and the mesenchymal stem cells under reduced oxygen tension.

14. The method of claim 13, wherein the neural stem cells and the mesenchymal stem cells are cultured in a culture medium comprising serum.

15. The method of claim 14, wherein the medical disorder is selected from the group consisting of ischemic stroke, Alzheimer'"'"'s disease, spinal cord injury, Parkinson'"'"'s disease, traumatic brain injury, solid tumors, multiple sclerosis, amyotrophic lateral sclerosis, Huntington'"'"'s disease, muscular dystrophy, retinitis pigmentosa, cerebral palsy, dementia, neuropathies due to chemotherapy or radiation therapy, and neuromyopathy.

16. The method of claim 14, wherein the medical disorder is ischemic stroke.

17. A kit for the treatment of a medical disorder comprising:

a. an effective amount of stem cell factors derived from neural stem cells;

b. an effective amount of stem cell factors derived from mesenchymal stem cells;

c. wherein the neural stem cells and mesenchymal stem cells are exposed to at least one environmental factor.

18. The kit of claim 17, wherein the at least one environmental factor comprises reduced oxygen tension.

19. The kit of claim 17, wherein the at least one environmental factor comprises culturing the neural stem cells and the mesenchymal stem cells under reduced oxygen tension.

20. The kit of claim 19, wherein the neural stem cells and the mesenchymal stem cells are cultured in a culture medium comprising serum.
