Abstract

The present invention provides novel microfluidic devices and methods that are useful for performing high-throughput screening assays and combinatorial chemistry. Such methods can include labeling a library of compounds by emulsifying aqueous solutions of the compounds and aqueous solutions of unique liquid labels on a microfluidic device, which includes a plurality of electrically addressable, channel bearing fluidic modules integrally arranged on a microfabricated substrate such that a continuous channel is provided for flow of immiscible fluids, whereby each compound is labeled with a unique liquid label, pooling the labeled emulsions, coalescing the labeled emulsions with emulsions containing a specific cell or enzyme, thereby forming a nanoreactor, screening the nanoreactors for a desirable reaction between the contents of the nanoreactor, and decoding the liquid label, thereby identifying a single compound from a library of compounds.

30. A method for manipulating droplets of a first fluid, the method comprising:

flowing droplets of the first fluid in a same direction in an immiscible continuous phase fluid within a microfluidic channel; and

adjusting spacing between the droplets by injecting a spacing fluid between the droplets or removing continuous phase fluid from between the droplets.

31. The method of claim 30, wherein the spacing fluid comprises the continuous phase fluid.

32. The method of claim 30, wherein removing continuous phase fluid comprises providing an outlet channel in fluid communication with the microfluidic channel constructed and arranged to allow passage of the continuous phase fluid and not the droplets.

33. The method of claim 30, further comprising the step of providing a second fluid in the microfluidic channel.

34. The method of claim 33, wherein the second fluid is a continuous stream.

35. The method of claim 33, wherein the second fluid comprises one or more droplets.

36. The method of claim 30, wherein the droplets are surrounded by the continuous phase fluid.

37. The method of claim 30, wherein the spacing fluid is injected from a direction substantially perpendicular to the microfluidic channel.

38. The method of claim 30, wherein the continuous phase fluid is removed substantially perpendicular to the microfluidic channel.

39. The method of claim 30, wherein the droplets are immiscible with the spacing fluid.

40. The method of claim 30, wherein the continuous phase fluid and the spacing fluid are different.

41. The method of claim 30, wherein the droplets have the same composition.

42. The method of claim 30, wherein the droplets have different compositions.

43. The method of claim 30, wherein the droplets contain a molecule selected from the group consisting of a nucleic acid, a cell, an enzyme, a substrate, an antibody, or a combination thereof.

44. The method of claim 43, wherein the droplets contain reagents for an amplification reaction.

45. The method of claim 30, wherein the continuous phase fluid is an oil.

46. The method of claim 45, wherein the oil comprises a surfactant.

47. The method of claim 46, wherein the surfactant is a fluorosurfactant.

48. The method of claim 30, wherein the continuous phase fluid is a fluorinated oil.

49. The method of claim 30, wherein the spacing fluid or continuous phase fluid is injected or removed using a syringe or pump.

50. The method of claim 30, wherein the spacing fluid or continuous phase fluid is injected or removed using positive or negative pressure source.

51. The method of claim 30, further comprising forming the droplets of the first fluid in an immiscible continuous phase fluid.
