Abstract

Methods and compositions are provided which comprise effective amounts of analgesic to treat a subject, including reducing or eliminating an adverse effect associated with the analgesic.

139. A solid composition, comprising an immediate-release layer that comprises an antiemetic, and a controlled-release layer that comprises an opioid analgesic and a non-opioid analgesic, wherein about 90% of the antiemetic is released in about 1 minute to about 20 minutes following contact of the solid composition with a dissolution fluid as measured by USP Apparatus 2 (Paddle Apparatus), and wherein 30-60% of the opioid analgesic is released in 5 to 10 minutes following contact of the solid composition with a dissolution fluid as measured by USP Apparatus 2 (Paddle Apparatus).

140. The solid composition of claim 139, wherein the opioid analgesic is hydrocodone, oxycodone, or a pharmaceutically acceptable salt thereof.

141. The solid composition of claim 140, wherein the hydrocodone or a pharmaceutically acceptable salt thereof is present in an amount of about 6.5-8.5 mg.

142. The solid composition of claim 141, wherein the hydrocodone or a pharmaceutically acceptable salt thereof is present in an amount of about 7.5 mg.

143. The solid composition of claim 142, wherein the pharmaceutically acceptable salt of the hydrocodone is hydrocodone bitartrate and is present in an amount of about 7.5 mg.

144. The solid composition of claim 141, wherein the hydrocodone is present in an amount of about 5 mg.

145. The solid composition of claim 140, wherein the opioid analgesic is hydrocodone bitartrate.

The solid composition of claim 140, wherein the oxycodone or a pharmaceutically acceptable salt thereof is present in an amount of about 0.5, 1.0, 1.5, 2.0, 2.5, 3.0, 3.5, 4.0, 4.5, 5.0, 5.5, 6.0, 6.5, 7.0, 7.5, 8.0, 8.5, 9.0, 9.5, 10.0, 10.5, 11.0, 11.5, 12.0, 12.5, 13.0, 13.5, 14.0, 14.5 or 15.0 mg.

147. The solid composition of claim 146, wherein the oxycodone or a pharmaceutically acceptable salt thereof is present in an amount of about 7.5 mg.

148. The solid composition of claim 146, wherein the oxycodone or a pharmaceutically acceptable salt thereof is present in an amount of about 10 mg.

149. The solid composition of claim 140, wherein the opioid analgesic is oxycodone hydrochloride.

150. The solid composition of claim 139, wherein the antiemetic is promethazine or a pharmaceutically acceptable salt thereof.

151. The solid composition of claim 150, wherein the promethazine or a pharmaceutically acceptable salt thereof is present in an amount of about 11-14 mg.

152. The solid composition of claim 151, wherein the promethazine or a pharmaceutically acceptable salt thereof is present in an amount of about 12.5 mg.

153. The solid composition of claim 152, wherein the pharmaceutically acceptable salt of the promethazine is promethazine hydrochloride and is present in an amount of about 12.5 mg.

154. The solid composition of claim 151, wherein the promethazine is present in an amount of about 11 mg.

155. The solid composition of claim 150, wherein the antiemetic is promethazine hydrochloride.

156. The solid composition of claim 139, wherein the solid composition is a bi-layer tablet.

157. The solid composition of claim 156, wherein the bi-layer tablet has a thickness of about 6.5 mm.

158. The solid composition of claim 156, wherein the bi-layer tablet has a hardness of about 15 kp.

159. The solid composition of claim 139, wherein the non-opioid analgesic is acetaminophen or a pharmaceutically acceptable salt thereof.

160. The solid composition of claim 159, wherein the acetaminophen or a pharmaceutically acceptable salt thereof is present in an amount of about 290-360 mg.

161. The solid composition of claim 160, wherein the acetaminophen or a pharmaceutically acceptable salt thereof is present in an amount of about 325 mg.

162. The solid composition of claim 139, wherein about 90% to about 100% of the opioid analgesic or non-opioid analgesic is released in about 40 minutes following contact of the solid composition with a dissolution fluid as measured by USP Apparatus 2 (Paddle Apparatus).
