Abstract

The invention provides isolated stem cells of non-embryonic origin that can be maintained in culture in the undifferentiated state or differentiated to form cells of multiple tissue types. Also provided are methods of isolation and culture, as well as therapeutic uses for the isolated cells.

Claims

64. A method for promoting wound healing in a subject, the method comprising administering the cells described below to the subject in a therapeutically effective amount and for a sufficient time to the wound to promote the wound healing in the subject:

(a) isolated expanded human non-embryonic, non-germ cells, the cells having undergone at least 10-40 doublings in culture, wherein the cells express oct4 and have a normal karyotype;

(b) isolated expanded human non-embryonic, non-germ cells that are obtained by culture of non-embryonic non-germ tissue, the cells having undergone 40 cell doublings in culture, wherein the cells have a normal karyotype;

(c) isolated expanded human multipotent non-embryonic non-germ cells that can differentiate into at least one cell type of at least two of the endodermal, ectodermal, and mesodermal embryonic lineages;

the cells having undergone at least 10-40 cell doublings in culture;

or

(d) isolated expanded human non-embryonic non-germ cells, the cells having undergone at least 10-40 cell doublings in culture, wherein the cells express telomerase and have a normal karyotype.

65. The method of claim 64(a) wherein the non-embryonic, non-germ cells further express one or more of telomerase, rex-1, rox-1, or sox-2.

66. The method of claim 64(a) wherein the non-embryonic, non-germ cells can differentiate into at least one cell type of at least two of the endodermal, ectodermal, and mesodermal embryonic lineages.

67. The method of claim 66 wherein the non-embryonic, non-germ cells further express one or more of telomerase, rex-1, rox-1, or sox-2.

68. The method of claim 66 wherein the non-embryonic, non-germ cells can differentiate into at least one cell type of each of the endodermal, ectodermal, and mesodermal embryonic lineages.

70. The method of claim 64(b) wherein the non-embryonic, non-germ cells express one or more of oet4, telomerase, rex-1, rox-1, or sox-2.

71. The method of claim 64(b) wherein the non-embryonic, non-germ cells can differentiate into at least one cell type of at least two of the endodermal, ectodermal, and mesodermal embryonic lineages.

72. The method of claim 71 wherein the non-embryonic, non-germ cells express one or more of oct4, telomerase, rex-1, rox-1, or sox-2.

73. The method of claim 71 wherein the non-embryonic, non-germ cells can differentiate into at least one cell type of each of the endodermal, ectodermal, and mesodermal embryonic lineages.

74. The method of claim 73 wherein the non-embryonic, non-germ cells express one or more of oct4, telomerase, rex-1, rox-1 or sox-2.

75. The method of claim 64(c) wherein the non-embryonic, non-germ cells express one or more of oct4, telomerase, rex-1, rox-1, or sox-2.

76. The method of claim 64(c) wherein the non-embryonic, non-germ cells can differentiate into at least one cell type of each of the endodermal, ectodermal, and mesodermal embryonic lineages.

77. The method of claim 76 wherein the non-embryonic, non-germ cells express one or more of oct4, telornerase, rex-1, rox-1, or sox-2.

78. The method of claim 64(d) wherein the non-embryonic, non-germ cells further express one or more of oct4, rex-1, rox-1, or sox-2.

79. The method of claim 64(d) wherein the non-embryonic, non-germ cells can differentiate into at least one cell type of at least two of the endodermal, ectodermal, and mesodermal embryonic lineages.

80. The method of claim 79 wherein the non-embryonic, non-germ cells further express one or more of oct4, rex-1, rex-1, or sox-2.

81. The method of claim 79 wherein the non-embryonic, non-germ cells can differentiate into at least one cell type of each of the endodermal, ectodermal, and mesodermal embryonic lineages.

82. The method of claim 81 wherein the non-embryonic, non-germ cells further express one or more of oct4, rex-1, rox-1, or sox-2.

83. The method of claim 64 wherein said cells are derived from bone marrow.

84. The method of claim 64 wherein the cells are administered by localized administration.

85. The method of claim 64 wherein the cells are administered by systemic administration.

86. The method of claim 64 wherein the cells are administered in a matrix.

87. The method of claim 64 wherein treatment results in the development of skin epithelial tissue.

88. The method of claim 64 wherein the cells are used in skin grafting.
