Abstract

Use of a composition for the prevention, amelioration and/or treatment of conditions, disorders and/or diseases associated with senescence and/or inflammation of the teeth and/or associated structures and tissues in the mouth, the composition comprising a conjugate of quinic acid with at least one molecule of caffeic acid, or a derivative, isomer or salt thereof.

Claims

1. Use of a composition for the prevention, amelioration and/or treatment of conditions, disorders and/or diseases associated with senescence and/or inflammation of the teeth and/or associated structures and tissues in the mouth, the composition comprising a conjugate of quinic acid with at least one molecule of caffeic acid, or a derivative, isomer or salt thereof.

2. Use of the composition of claim 1, wherein the conjugate is represented by the following formula (I):

3. Use of the composition of claim 1 or 2, wherein the conjugate is an ester of caffeic acid and quinic acid.

4. Use of the composition of any preceding claim, wherein the composition comprises a dicaffeoylquinic acid and/or a tricaffeoylquinic acid, or a derivative, isomer or salt thereof.

Use of the composition of any preceding claim, wherein the conjugate is selected from 1,3-dicaffeoylquinic acid, 1,4-dicaffeoylquinic acid, 1,5-dicaffeoylquinic acid, 3,4-dicaffeoylquinic acid, 3,5-dicaffeoylquinic acid, 4,5-dicaffeoylquinic acid, or 3,4,5-tricaffeoylquinic acid, or a derivative, isomer or salt thereof.

6. Use of the composition of any preceding claim, wherein the composition inhibits one or more of the matrix metalloproteinases (MMPs), nuclear factor kappa B, cytokines and/or tissue inhibitors of matrix metalloproteinase (TIMP) activators.

7. Use of the composition of any preceding claim for the prevention, amelioration and/or treatment of disorders and/or diseases associated with an increase in the expression of one or more matrix metalloproteinase (MMP) genes and/or proteins.

8. Use of the composition of any preceding claim, wherein the composition inhibits the activity of MMP-9 and/or MMP-2.

9. Use of the composition of any preceding claim, wherein the composition enhances extracellular matrix cohesion of the teeth and associated structures and tissues of the mouth.

10. Use of the composition of any preceding claim, wherein the composition affects the concentration of MMP-9 in saliva.

11. Use of the composition of any preceding claim, wherein the composition activates the expression of the sirtuin 3 gene to enhance the production of the sirtuin 3 protein and/or inhibits the expression of the sirtuin 2 gene to inhibit the production of the sirtuin 2 protein.

12. Use of the composition of any preceding claim for the prevention, amelioration and/or treatment of demineralisation of teeth and/or associated structures in the mouth.

13. Use of the composition of any preceding claim for the prevention, amelioration and/or treatment of demineralisation of the enamel and/or dentine of the teeth.

14. Use of the composition of any preceding claim for the stimulation and/or promotion of remineralisation of teeth and/or associated structures in the mouth following demineralisation.

15. Use of the composition of any preceding claim for the stimulation and/or promotion of the remineralisation of the enamel and/or dentine of the teeth.

16. Use of the composition of any preceding claim for the prevention, amelioration and/or treatment of leukoplakia.

17. Use of the composition of any preceding claim for the prevention, amelioration and/or treatment of chronic inflammatory disease of the teeth and associated structures of the mouth.

18. Use of composition of any preceding claim for prevention, amelioration and/or treatment of conditions of the teeth and/or associated structures in the mouth associated with senescence.

19. Use of the composition of any preceding claim for the prevention, amelioration and/or treatment of tissue shrinkage or contraction which may occur following application of a dental implant or prosthesis in the mouth.

20. Use of the composition of any preceding claim wherein the composition may be incorporated into a coating or other material used in a dental implant or prosthesis.

21. Use of the composition of any preceding claim for the prevention, amelioration and/or treatment of dental caries.

22. Use of the composition of any preceding claim for incorporation into teeth whitening agents.

23. Use of the composition of claim 22 for the prevention, amelioration and/or treatment of conditions affected by free radicals, hydrogen peroxide and/or associated derivatives which may be generated by the use of such agents.

24. Use of the composition of any preceding claim, wherein the conjugate may be extracted from a natural source.

25. Use of the composition of claim 24, wherein the conjugate is a plant extract.

26. Use of the composition of claim 25, wherein the conjugate is an artichoke extract.

27. Use of the composition of claim 24, wherein the conjugate may be synthesised or genetically engineered from plants, bacteria, fungi, mould, algae or another suitable source.

28. Use of the composition of any preceding claim, wherein the conjugate comprises at least one acceptable carrier and may be carried within a vesicle, micelle, liposome, nanoparticle or other suitable vehicle, including dental inserts.

29. Use of the composition of any preceding claim, wherein the composition is directly applied to the teeth or associated structures in the mouth.

30. Use of the composition of claim 29, wherein the composition is directly applied to the teeth or associated structures in the mouth by a dental practitioner during a surgical procedure, or by injection.

31. Use of the composition of any preceding claim, wherein the composition is applied topically to the teeth and/or associated structures.

32. Use of the composition of claim 31, wherein the composition is applied in the form of a mouthwash, paste such as toothpaste, lotion, gel, powder or other suitable medium.

33. Use of the composition of any preceding claim, wherein the conjugate is provided in the form of a free base or a pharmaceutically acceptable salt.

34. Use of the composition of any preceding claim, wherein the composition is used in combination with another therapy, including antibodies, analgesics, substances used to neutralise the acidity of saliva, chemotherapy, synergistic compounds, genes, gene products, cell therapy and/or radiation.

35. Use of a composition as defined herein and with reference to the accompanying drawings.
