Abstract

The present invention relates to systems that utilize a combination of immunoassay and magnetic immunoassay techniques to detect an analyte within an extended range of specified concentrations. In particular, a device includes a first electrochemical sensor positioned within a conduit adjacent to a second electrochemical sensor and spaced apart from one another at a predetermined distance. The first electrochemical sensor includes an immobilized layer of antibodies. The second electrochemical sensor includes a magnetic field disposed locally around the second electrochemical sensor, and the magnetic field is configured to attract magnetic beads onto a surface of the second electrochemical sensor. The device further includes a scavenging electrode positioned between the first electrochemical sensor and the second electrochemical sensor. The scavenging electrode is configured to prevent crosstalk between the first electrochemical immunosensor and the second electrical immunosensor.

Claims

1. A device for detecting an analyte in a biological sample comprising:

a housing including a conduit;

a first electrochemical sensor positioned within the conduit, wherein the first electrochemical sensor including an immobilized layer of antibodies configured to bind to a first complex of signal antibodies and the analyte to form a second complex;

a second electrochemical sensor positioned within the conduit and adjacent to the first electrochemical sensor, wherein the second electrochemical sensor includes a magnetic field disposed locally around the second electrochemical sensor, and the magnetic field is configured to attract magnetic beads onto a surface of the second electrochemical sensor such that a third complex of the first complex and antibodies immobilized on the magnetic beads is localized on the second immunosensor; and

a scavenging electrode positioned between the first electrochemical sensor and the second electrochemical sensor,

wherein the first electrochemical sensor and the second electrochemical sensor are spaced apart from one another at a predetermined distance.

2. The device of claim 1, wherein the first electrochemical sensor and the second electrochemical sensor are spaced apart from one another at a predetermined distance of at least 0.2 mm.

The device of claim 1, further comprising a conductivity sensor configured to determine a position of the biological sample on the substrate with respect to the first electrochemical sensor and the second electrochemical sensor, wherein the scavenging electrode is connected to the conductivity sensor.

4. The device of claim 1, wherein the scavenging electrode is configured to prevent electroactive species generated in a region of the second electrochemical sensor from diffusing to the first sensor.

5. The device of claim 1, wherein the scavenging electrode is configured to prevent electroactive species generated in a region of the second electrochemical sensor from being transported to the first electrochemical sensor.

6. The device of claim 1, wherein the scavenging electrode is configured to prevent electroactive species generated in a region of the second electrochemical sensor from being detected at the first electrochemical sensor.

7. The device of claim 1, wherein the scavenging electrode is configured to prevent electroactive species generated in a region of the first electrochemical sensor from diffusing to the second sensor.

8. The device of claim 1, wherein the scavenging electrode is configured to prevent electroactive species generated in a region of the first electrochemical sensor from being transported to the second electrochemical sensor.

9. The device of claim 1, wherein the scavenging electrode is configured to prevent electroactive species generated in a region of the first electrochemical sensor from being detected at the second electrochemical sensor.

10. The device of claim 1, further comprising a first reagent region coated with the signal antibodies for the analyte.

11. The device of claim 10, further comprising a second reagent region coated with the magnetic beads.

12. The device of claim 11, wherein the first electrochemical sensor, the second electrochemical sensor, the first reagent region, and the second reagent region are located on a sensor chip.

13. The device of claim 1, further comprising a composite material including a binder and a particulate magnetic material, the particulate magnetic material configured to generate the magnetic field.

14. The device of claim 13, wherein the binder is a polyimide or polyvinyl alcohol (PVA).

15. The device of claim 13, wherein the particulate magnetic material is comprised of neodymium iron boron (NdFeB) alloy or aluminum nickel cobalt (AlNiCo) alloy.

16. The device of claim 1, wherein the signal antibodies are conjugated with an enzyme.

17. A device comprising;

a conduit;

a substrate including a planar top and bottom surface positioned within the conduit;

a first electrochemical sensor positioned on the top surface of the substrate, the first electrochemical sensor including an immobilized layer of antibody configured to bind to an analyte;

a second electrochemical sensor positioned on the top surface of the substrate;

an opening in the bottom surface of the substrate extending to a region in the substrate below the second electrochemical sensor;

a composite material including a binder and a particulate magnetic material;

a first dry reagent positioned with the conduit, the first dry reagent comprising magnetic beads including an immobilized layer of antibody configured to bind to the analyte;

a second dry reagent positioned with the conduit, the second dry reagent comprising signal antibodies configured to bind to the analyte; and

a scavenging electrode positioned between the first electrochemical sensor and the second electrochemical sensor,

wherein the first electrochemical sensor and the second electrochemical sensor are spaced apart from one another at a predetermined distance of at least 0.2 mm, and

wherein the particulate magnetic material generates a magnetic field that is aligned with respect to the second electrochemical sensor.

18. A device for detecting an analyte in a biological sample comprising:

a first electrochemical sensor formed on a substrate, the first electrochemical sensor including an immobilized layer of antibody configured to bind to the analyte;

a second electrochemical sensor formed on the substrate and spaced a predetermined distance from the first electrochemical sensor;

an opening in the bottom surface of the substrate extending to a region in the substrate aligned vertically with respect to the second electrochemical sensor;

a composite material including a particulate magnetic material substantially filling the opening;

magnetic beads including an immobilized layer of antibody configured to bind to the analyte; and

a scavenging electrode positioned between the first electrochemical sensor and the second electrochemical sensor,

wherein the first electrochemical sensor and the second electrochemical sensor are spaced apart from one another at a predetermined distance of at least 0.2 mm, and

wherein the particulate magnetic material generates a magnetic field that is vertically aligned with respect to the second electrochemical sensor.

19. The device of claim 18, wherein the scavenging electrode is connected to the second electrochemical sensor.

20. The device of claim 18, further comprising a conductivity sensor configured to determine a position of the biological sample on the substrate with respect to the first electrochemical sensor and the second electrochemical sensor, wherein the scavenging electrode is connected to the conductivity sensor.
