1. A method for the analysis of an analyte in a test solution comprising the steps of:
- (a) non-diffusively immobilizing in each of a predetermined number of successive, spaced reaction zones of a liquid permeable medium a reactant capable of reacting with the analyte to form a predetermined product;
(b) flowing said solution along the medium and sequentially through the reaction zones; and
(c) detecting the presence of analyte, said reactant or said predetermined product in the reaction zones, wherein the number of zones in which detection occurs is related to the amount of analyte in the solution.

Abstract
A method and apparatus for the quantitative determination of an analyte in a liquid employs a liquid-permeable solid medium defining a liquid flow path. The medium includes a number of reaction-containing reaction zones spaced apart along the flow path and in which reaction occurs with the analyte or an analyte derivative (e.g., a labeled analyte) to result in the formation of a predetermined product. Detector means are employed to detect analyte, analyte derivatives, reactant or predetermined product in the reaction zones, the number of such zones in which such detection occurs indicating the amount of analyte in the liquid.

20. A method for the quantitative analysis of an analyte which is a member of a ligand-antiligane binding pair in a test solution comprising the steps of:

(a) providing a liquid-permeable solid medium defining a flow path and having a predetermined number of successive, spaced reaction zones in the path of flow, the reaction zones having non-diffusively immobilized therein the other member of said binding pair that is capable of binding to the analyte to form a predetermined product; flowing said solution along the flow path and sequentially through the spaced reaction zone; and detecting the presence of analyte in the reaction zones, wherein the number of zones in which such detection occurs is related to the amount of analyte in the solution. - View Dependent Claims (21)

26. A method for the analysis of an analyte which is a member of a ligand-antiligand binding pair in a test solution comprising the steps of:

providing a liquid-permeable solid medium defining a flow path for the movement of the analyte and having a zone for the application of the solution to said medium and at least one reaction zone;
said reaction zone having non-diffusively immobilized therein a predetermined amount of a reactant that is the other of said ligand-antiligand binding pair and that is capable of binding to the analyte to form a predetermined product;reacting the analyte, said reactant or said predetermined product with a second ligand-antiligand binding pair member, wherein said second ligand-antiligand binding pair member is attached to a detectable label; flowing said solution along the flow path and sequentially through the application zone and the reaction zone; and detecting the presence of said label in the reaction zone, wherein detection occurs in the reaction zone only if analyte is present in the test solution in a predetermined amount and wherein said detecting step comprises detecting the presence of said label.
