Abstract

Methods for conducting controlled environment electrotransport are described. The methods generally concern control of the ionic environment of the active electrode reservoir, in an electrotransport system. This control may be utilized, for example, to maintain a constant extraneous ion presence, such as pH. It may also be applied in a preferred manner to maintain selected control over the delivery rate of a target species, in time, in the presence of extraneous ions. Apparatus usable to provide for controlled environment electrotransport are also described. In general the apparatus comprises an electrotransport system including an active electrode reservoir having therein both a primary electrode arrangement and a secondary electrode arrangement, with a control arrangement for selected operation of both. The secondary electrode arrangement, being operated different from the primary electrode arrangement, is selected for operation to selectively enhance or otherwise modify ion presence in the active electrode reservoir, to achieve application of the general methods described. The apparatus may include a sensor means therein, for use as feedback to a control system during operation. Methods and systems involving diffusion type delivery are also described.

Claims

1. A method of controlling delivery of a species, having a polar form and a less polar form, from a reservoir;

said method comprising the steps of;

(a) providing first and second electrode arrangements in electrically conductive contact with the reservoir; and

,

(b) selectively operating the first and second electrodes to control pH of the reservoir in a manner obtaining selected relative concentrations of the polar and less polar forms of the species, to obtain selected delivery.

2. The method according to claim 1 wherein the polar form is charged and the less polar form is uncharged.

3. A method according to claim 1 wherein the polar form of the species generated during the method is a protonated base.

4. An electrotransport apparatus comprising:

(a) an active electrode reservoir including a concentration of target species and a concentration of hydronium ions;

(b) a primary electrode arrangement, in said active electrode reservoir, having means for adjusting a rate of transport of the target species from the active electrode reservoir;

(c) a secondary electrode system, in said active electrode reservoir, having means for adjusting hydronium ion content in said reservoir; and

,

(d) means for operating said secondary electrode system to achieve maintenance of a constant hydronium ion concentration throughout operation of said apparatus.

5. An electrotransport apparatus comprising:

(a) an active electrode reservoir including a concentration of target ion species and a concentration of hydroxyl ions;

(b) a primary electrode arrangement, in said active electrode reservoir, having means for adjusting a rate of transport of the target species from the active electrode reservoir;

(c) a secondary electrode system, in said active electrode reservoir, having means for adjusting hydroxyle ion content in said reservoir; and

,

(d) means for operating said secondary electrode system to achieve maintenance of a constant hydroxyl ion concentration throughout operation of said apparatus.

6. An electrotransport apparatus comprising:

(a) an active electrode reservoir;

(b) a primary electrode arrangement in said active electrode reservoir;

(c) a secondary electrode arrangement in said active electrode reservoir;

said secondary electrode arrangement being isolated from direct contact with said primary electrode arrangement;

(d) at least one therapeutic species in said active electrode reservoir;

(e) said primary electrode arrangement being selectively operable to provide electromotive force to transport therapeutic ions from said active electrode reservoir; and

,

(f) said secondary electrode arrangement being selectively operable to generate electrochemical reactions in said active electrode reservoir ouch that an effect of the electrochemical reactions generated by the secondary electrode arrangement on an ionic content of the active electrode reservoir is different from that of the primary electrode arrangement, to selectively effect relative concentrations of ions in the active electrode reservoir.

7. An apparatus according to claim 6 wherein:

(a) said primary electrode arrangement is an anode; and

,

(b) said secondary electrode arrangement is an anode.

8. An apparatus according to claim 6 wherein:

(a) said active electrode reservoir includes material for generation of at least one therapeutic cation species and at least one extraneous cation species, upon operation of said primary electrode arrangement and said secondary electrode arrangement;

(b) said primary electrode arrangement being operable to transport said at least one therapeutic cation species and said at least one extraneous cation species, from said active electrode arrangement; and

,

(c) said secondary electrode arrangement being operable to introduce said at least one extraneous cation species into said active electrode reservoir.
