Abstract

The present invention provides an analytical rotor which holds a fluid in a chamber, typically to allow mixing with a reagent, and transfers the fluid to a receiving chamber in the rotor. The rotor comprises a holding chamber having an single exit duct and a receiving chamber connected to the holding chamber through the exit duct. The exit duct prevents flow at a first rotational speed and allows flow at a second, higher rotational speed. The exit duct may include a capillary passage in which capillary forces prevent flow until the rotational speed is increased. Alternatively, the exit duct may include a siphon.

Claims

1. An analytical rotor comprising:

a delivery chamber containing a fluid;

a holding chamber positioned radially outward from the delivery chamber, the holding chamber being connected to the delivery chamber through an entry channel, whereby the fluid flows from the delivery chamber to the holding chamber as the rotor is spinning at a first rotational speed;

a receiving chamber positioned radially outward from the holding chamber and being connected to the holding chamber through an exit duct; and

a capillary passage connecting the exit duct to the receiving chamber, the capillary passage having a cross sectional area less than the cross sectional area of the entry channel which prevents flow of fluid from the holding chamber to the receiving chamber when the rotor is spinning at the first rotational speed and allows flow at a second higher rotational speed.

2. The rotor of claim 1, wherein the holding chamber comprises a reagent.

3. The rotor of claim 2, wherein the reagent is coated on a solid surface in the holding chamber.

4. The rotor of claim 3, wherein the solid surface is a bead disposed within the holding chamber.

5. The rotor of claim 2, wherein the reagent is a marking compound.

6. The rotor of claim 1, wherein the fluid is a biological fluid.

7. The rotor of claim 1, wherein the fluid is a diluent.

8. The rotor of claim 1, wherein the receiving chamber is a separation chamber having a cell trap.

9. The rotor of claim 8, further comprising a collection chamber connected to the separation chamber and having a plurality of cuvettes disposed radially outward from the collection chamber.

10. The rotor of claim 1 wherein the fluid is blood.

11. The rotor of claim 1 wherein the first rotational speed is sufficient to generate a centrifugal force of about 20×

g to 27×

g.

12. The rotor of claim 1 wherein the second rotational speed is sufficient to generate a centrifugal force of about 45×

g.

13. The rotor of claim 1 wherein the delivery chamber contains a diluent.

14. A centrifugal rotor comprising:

a holding chamber comprising reagent applied to a solid surface in the holding chamber and having an exit duct,

wherein said solid surface is a bead disposed within the holding chamber,

a separation chamber, having a cell trap, positioned radially outward from the holding chamber and being connected to the holding chamber through the exit duct; and

a siphon connecting the exit duct to the separation chamber.

15. The rotor of claim 14, wherein the reagent is a marking compound.

16. The rotor of claim 14, further comprising a collection chamber connected to the separation chamber and having a plurality of cuvettes disposed radially outward from the collection chamber.

17. The rotor of claim 14, further comprising siphon connecting an exit duct in the separation chamber to a receiving chamber.

18. A centrifugal rotor comprising:

a holding chamber having an exit duct;

a metering chamber connected to an overflow chamber, the metering chamber being positioned radially outward from the holding chamber and being connected to the holding chamber through the exit duct; and

a siphon connecting the exit duct to the metering chamber.
