Abstract

The present invention is a system for chemometric analysis for the extraction of the individual component fluorescence spectra and fluorescence lifetimes from a target mixture. The present invention combines a processor with an apparatus for generating an excitation signal to transmit at a target mixture and an apparatus for detecting the emitted signal from the target mixture. The present invention extracts the individual fluorescence spectrum and fluorescence lifetime measurements from the frequency and wavelength data acquired from the emitted signal. The present invention uses an iterative solution that first requires the initialization of several decision variables and the initial approximation determinations of intermediate matrices. The iterative solution compares the decision variables for convergence to see if further approximation determinations are necessary. If the solution converges, the present invention then determines the reduced best fit error for the analysis of the individual fluorescence lifetime and the fluorescence spectrum before extracting the individual fluorescence lifetime and fluorescence spectrum from the emitted signal of the target mixture.

Claims

1. A method of chemometric analysis for the extraction of an individual fluorescence spectrum and an individual fluorescence lifetime from a target mixture comprising the following steps:

acquiring the frequency and wavelength data from the emitted signal from the target mixture;

determining a G matrix and a S matrix from a frequency vector and a wavelength vector;

renormalizing w^-1 S_x to G_x ;

determining an initial U, V, g and w^-1 s approximations;

initializing one or more decision variables;

comparing the decision variables for convergence to see if further determinations for U, V, g and w^-1 s are necessary;

extracting the individual fluorescence lifetime from the target mixture; and

extracting the individual fluorescence spectrum from the target mixture.

2. The method of claim 1 wherein the step of determining the initial U, V, g and w^-1 s approximations further comprises the following steps:

determining the initial U approximation;

determining the initial V approximation; and

determining the initial g and w^-1 s approximations.

3. The method of claim 1 wherein the step of comparing the decision variables for convergence further comprises the following steps:

determining a next U approximation;

determining a next V approximation;

determining a next g and w^-1 s approximations; and

updating the decision variables.

4. The method of claim 1 further comprising the step of determining a best fit error for an analysis of the individual fluorescence lifetime and the fluorescence spectrum.

A program storage device readable by a computer, tangibly embodying a program of instructions executable by the computer to perform method steps for a method of chemometric analysis for the extraction of an individual fluorescence spectrum and an individual fluorescence lifetime from a target mixture, comprising the following method steps:

acquiring the frequency and wavelength data from the emitted signal from the target mixture;

determining a G matrix and a S matrix from a frequency vector and a wavelength vector;

renormalizing w^-1 S_x to G_x ;

determining an initial U, V, g and w^-1 s approximations;

initializing one or more decision variables;

comparing the decision variables for convergence to see if further determinations for U, V, g and w^-1 s are necessary;

extracting the individual fluorescence lifetime from the mixture; and

extracting the individual fluorescence spectrum from the target mixture.

6. The program storage device of claim 5 wherein said step of determining the initial U, V, g and w^-1 s approximations further comprise the following steps:

determining the initial U approximation;

determining the initial V approximation; and

determining the initial g and w^-1 s approximations.

7. The program storage device of claim 5 wherein said step of comparing the decision variables for convergence further comprises the following steps:

determining a next U approximation;

determining a next V approximation;

determining a next g and w^-1 s approximations; and

updating the decision variables.

8. The program storage device of claim 5 further comprising the step of determining a best fit error for an analysis of the individual fluorescence lifetime and the fluorescence spectrum.

9. A system for chemometric analysis for the extraction of an individual fluorescence spectrum and an individual fluorescence lifetime from a target mixture, comprising:

a processor that acquires the frequency and wavelength data from the emitted signal from the mixture;

means for determining a G matrix and a S matrix from a frequency vector and a wavelength vector;

means for renormalizing w^-1 S_x to G_x ;

means for determining an initial U, V, g and w^-1 s approximations;

means for initializing one or more decision variables;

means for comparing the decision variables for convergence to see if further determinations for U, V, g and w^-1 s are necessary;

means for extracting the individual fluorescence lifetime from the target mixture; and

means for extracting the individual fluorescence spectrum from the target mixture.

10. The system of claim 9 wherein said means for determining the initial U, V, g and w^-1 s approximations further comprise:

means for determining the initial U approximation;

means for determining the initial V approximation; and

means for determining the initial g and w^-1 s approximations.

11. The system of claim 9 wherein said means for comparing the decision variables for convergence further comprises:

means for determining a next U approximation;

means for determining a next V approximation;

means for determining a next g and w^-1 s approximations; and

means for updating the decision variables.

12. The system of claim 9 further comprising means for determining a best fit error for an analysis of the individual fluorescence lifetime and the fluorescence spectrum.

13. An apparatus for chemometric analysis for the extraction of an individual fluorescence spectrum and an individual fluorescence lifetime from a target mixture, comprising:

a processor that acquires the frequency and wavelength data from the emitted signal from the mixture;

a determiner that determines a G matrix and a S matrix from a frequency vector and wavelength vector;

a renormalizer that renormalizes w^-1 S_x to G_x ;

a determiner that determines an initial U, V, g and w^-1 s approximations;

an initializer that initializes one or more decision variables;

a comparator that compares the decision variables for convergence to see if further determinations for U, V, g and w^-1 s are necessary;

an extractor that extracts the individual fluorescence lifetime from the target mixture; and

an extractor that extracts the individual fluorescence spectrum from the target mixture.

14. The method of claim 13 wherein said determiner that determines the initial U, V, g and w^-1 s approximations further comprises:

a determiner that determines the initial U approximation;

a determiner that determines the initial V approximation; and

a determiner that determines initial g and w^-1 s approximations.

15. The method of claim 13 wherein said comparator that compares the decision variables for convergence to see if further determinations are necessary further comprises:

a determiner that determines a next U approximation;

a determiner that determines a next V approximation;

a determiner that determines a next g and w^-1 s approximations; and

an updater that updates the decision variables.

16. The method of claim 13 further comprising a determiner that determines a best fit error for an analysis of the individual fluorescence lifetime and the fluorescence spectrum.
