Abstract

The present invention provides a microfabricated sensor and a method capable of rapid simultaneous measurement of multiple analytes in a fluid sample. The sensor is inexpensive, disposable and portable, and requires only microliters of sample, a particular advantage with precious fluids such as blood. The sensor utilizes diffusion between layered laminar streams rather than side by side streams. This allows multiple side by side channels for simultaneous detection of multiple analytes. In the sensor, a sample stream and a carrier stream flow in layers, one on top of the other, and one or more reagents are introduced to the bottom of the carrier stream through either a fluid or a solid reagent inlet. The reagent contains reagent particles which, in the presence of the analyte, have a detectable change in a property. The analyte diffuses into the carrier stream where it interacts with reagent particles and is detected by optical, electrochemical or other devices.

Claims

1. A diffusion based sensor, comprising:

a sample inlet;

an outlet;

a laminar flow channel connected to said sample inlet and said outlet, whereby a laminar flow of sample fluid can be established between said sample inlet and said outlet;

a carrier inlet connecting with the bottom of said laminar flow channel between said sample inlet and said outlet, whereby a laminar flow of carrier fluid can be established between said carrier inlet and said outlet, said carrier fluid forming a layer below said sample fluid;

said channel being greater in depth at said carrier inlet than at said sample inlet;

a first reagent inlet connecting with the bottom of said channel between said carrier inlet and said outlet.

2. The sensor of claim 1 further comprising a second reagent inlet connecting with the bottom of said channel between said carrier inlet and said outlet.

3. The sensor of claim 2 wherein said first and second reagent inlets are positioned in series.

4. The sensor of claim 1 wherein the depth of said channel increases at said carrier inlet.

5. The sensor of claim 1 wherein said channel is a convoluted channel.

6. The sensor of claim 1 wherein the length of said channel is between about 5 mm and 50 mm.

7. The sensor of claim 1 wherein the width of said channel is between 0.1 and 1 cm.

8. The sensor of claim 1 wherein the depth of said channel is between 5 and 500 μ

m.

9. The sensor of claim 8 wherein the depth of said channel is between 50 and 100 μ

m.

10. The sensor of claim 1 wherein said first reagent inlet is a fluid inlet.

11. The sensor of claim 1 wherein said first reagent inlet is a solid inlet.

12. The sensor of claim 11 wherein said solid inlet is a cavity in said channel.

13. The sensor of claim 1 wherein said sample inlet is preceded by a filter.

14. The sensor of claim 13 wherein said filter is a diffusion based microfilter.

15. The sensor of claim 14 wherein said microfilter comprises:

a filter laminar flow channel having an upstream end and a downstream end, and connected on said downstream end to said sample inlet;

an extraction stream inlet connected to said laminar flow channel on said upstream end;

an unfiltered-sample stream inlet connected to said laminar flow channel on said upstream end; and

a residual-sample stream outlet connected to said laminar flow channel on said downstream end.

16. The sensor of claim 1 for use with optical detection wherein at least a portion of said channel is formed in a transparent material.

17. The sensor of claim 16 wherein said transparent material is plastic.

18. The sensor of claim 16 wherein said channel is formed in a silicon plate with a transparent cover plate.

19. The sensor of claim 1 for use with electrochemical detection further including an electrode positioned in said channel between said reagent inlet and said outlet.

20. The sensor of claim 19 further including one or more additional electrodes positioned in said channel between said reagent inlet and said outlet.

21. The sensor of claim 20 wherein said electrodes are positioned in series.

22. The sensor of claim 19 wherein said electrode is deposited on the surface of said channel.

23. The sensor of claim 19 for use with both electrochemical and optical detection, wherein at least a portion of said channel is formed in a transparent material.

24. A diffusion based sensor, comprising:

a sample inlet;

an outlet;

a laminar flow channel connected to said sample inlet and said outlet, whereby a laminar flow of sample fluid can be established between said sample inlet and said outlet;

a carrier inlet connecting with the bottom of said laminar flow channel between said sample inlet and said outlet, whereby a laminar flow of carrier fluid can be established between said carrier inlet and said outlet, said carrier fluid forming a layer below said sample fluid;

a first reagent inlet connecting with the bottom of said channel between said carrier inlet and said outlet;

a second reagent inlet connecting with the bottom of said channel between said carrier inlet and said outlet;

wherein said first and second reagent inlets are positioned in parallel, and wherein the width of each of said reagent inlets is less than the width of said channel.

25. A diffusion based sensor, comprising:

a sample inlet;

an outlet;

a laminar flow channel connected to said sample inlet and said outlet, whereby a laminar flow of sample fluid can be established between said sample inlet and said outlet;

a carrier inlet connecting with the bottom of said laminar flow channel between said sample inlet and said outlet, whereby a laminar flow of carrier fluid can be established between said carrier inlet and said outlet, said carrier fluid forming a layer below said sample fluid;

a first, solid, reagent inlet formed by a cavity in said channel connecting with the bottom of said channel between said carrier inlet and said outlet;

said solid inlet further including a reagent pellet immobilized in said cavity.

26. The sensor of claim 25 wherein said pellet comprises reagent particles in a soluble reagent carrier.

27. A diffusion based sensor, comprising:

a sample inlet;

an outlet;

a laminar flow channel connected to said sample inlet and said outlet, whereby a laminar flow of sample fluid can be established between said sample inlet and said outlet;

a carrier inlet connecting with the bottom of said laminar flow channel between said sample inlet and said outlet, whereby a laminar flow of carrier fluid can be established between said carrier inlet and said outlet, said carrier fluid forming a layer below said sample fluid;

means for injecting fluid into the device in fluid communication with said carrier inlet;

a first reagent inlet connecting with the bottom of said channel between said carrier inlet and said outlet.
