Abstract

The invention relates to nucleic acids covalently coupled to electrodes via conductive oligomers. More particularly, the invention is directed to the site-selective modification of nucleic acids with electron transfer moieties and electrodes to produce a new class of biomaterials, and to methods of making and using them.

Claims

1. A method for attaching a conductive oligomer to a gold electrode comprising:

(a) providing a conductive olizomer with at least a first subunit comprising a sulfur atom protected by a protective group selected from the group consisting of ethyl pyridine or trimethylsilylethyl; and

(b) attaching said sulfur atom to said gold electrode.

2. A method according to claim 1, further comprising adding additional subunits to form said conductive oligomer.

3. A method according to claim 2, further comprising adding at least a first nucleoside to said conductive oligomer.

4. A method according to claim 3, further comprising adding additional nucleosides to said first nucleoside to form a nucleic acid.

5. A method according to claim 1 wherein said protecting group is removed prior to said attachment.

6. A method for attaching a conductive oligomer to a gold electrode comprising:

a) providing a conductive oligomer having a covalently attached nucleic acid and a sulfur atom protected by a protecting group; and

b) attaching said sulfur atom to said gold electrode.

7. A method according to claim 6 wherein said protecting group is ethyl pyridine.

8. A method according to claim 6 wherein said protecting group is trimethylsilylethyl.

9. A method of attaching a composition to a gold electrode comprising:

a) providing a first monolayer forming species comprising a covalently attached nucleic acid and a sulfur atom protected by a protecting group;

b) providing a conductive oligomer comprising at least a first subunit with a sulfur atom protected by a protecting group; and

c) attaching said first and second species to said gold electrode.

10. A method according to claim 9 further comprising:

a) providing a third species comprising an insulator; and

b) attaching said third species to said gold electrode.

11. A method according to claim 9 or 10 wherein said protecting group is selected from ethyl pyridine or trimethylsilylethyl.
