Abstract

The invention relates generally to methods and apparatus for conducting analyses, particularly microfluidic devices for the detection of target analytes.

Claims

1. A microfluidic device comprising:

a substrate comprising wells in fluid communication with a flow channel, said flow channel capable of receiving fluid flow through an inlet port;

a population of beads, wherein said population comprises beads having a binding ligand, wherein the beads are present in said wells, wherein a well comprises at least a first bead of said population and a second bead of said population, and wherein said first bead comprises a nucleic acid and said second bead comprises an enzyme; and

an electroosmotic pump in fluid communication with said flow channel, wherein said electroosmotic pump is configured to deliver a fluid through said flow channel.

2. The microfluidic device of claim 1, wherein said binding ligand comprises a peptide.

3. The microfluidic device of claim 1, wherein said binding ligand comprises a nucleic acid.

4. The microfluidic device of claim 3, wherein said nucleic acid comprises a primer.

5. The microfluidic device of claim 4 further comprising an enzyme for extension of said primer.

6. The microfluidic device of claim 1, wherein one or more of said wells comprise a single bead.

7. The microfluidic device of claim 1, wherein said first bead and said second bead are different sizes.

8. The microfluidic device of claim 1, wherein said substrate comprises plurality of flow channels.

9. The microfluidic device of claim 8, wherein said flow channels are parallel to each other.

10. The microfluidic device of claim 1, wherein said substrate is configured for multiple samples.

11. The microfluidic device of claim 1, wherein said substrate is a chargeable substrate.

12. The microfluidic device of claim 1, wherein said flow channel comprises a fluid having ions dissolved therein.

13. The microfluidic device of claim 1, wherein said flow channel comprises an outlet port.

14. The microfluidic device of claim 1, wherein said electroosmotic pump is present on said substrate.

15. The microfluidic device of claim 1, wherein said electroosmotic pump is separate from said substrate.

16. A method of sequencing a nucleic acid, said method comprising:

providing a fluid comprising a target nucleic acid to the electroosmotic pump of the device of claim 4;

amplifying said target nucleic acid, thereby generating an amplified target nucleic acid; and

sequencing said amplified target nucleic acid.

17. The method of claim 16, wherein sequencing said amplified target nucleic acid further comprises providing to said electroosmotic pump a fluid comprising a primer complementary to said amplified target nucleic acid, a labeled terminator nucleotide and a polymerase enzyme.

18. A method of detecting a target analyte in a fluid, said method comprising:

providing a fluid comprising a target analyte to the electroosmotic pump of the device of claim 1;

allowing said target analyte to bind to said binding ligand; and

detecting the target analyte bound to said binding ligand.

19. The method of claim 18, wherein said target analyte comprises a nucleic acid.

20. The method of claim 19, wherein said binding ligand comprises a nucleic acid complementary to said nucleic acid.

21. The method of claim 18, wherein detecting the target nucleic acid bound to said binding ligand comprises detecting an optical signal.

22. The microfluidic device of claim 1, wherein the first bead and second bead are randomly distributed in said well.
