Abstract

The present invention is directed to devices, systems and methods for removing undesirable materials from a sample fluid by contact with a second fluid. The sample fluid flows as a thin layer adjacent to, or between, concurrently flowing layers of the second fluid, without an intervening membrane. In various embodiments, a secondary separator is used to restrict the removal of desirable substances and effect the removal of undesirable substances from blood. The invention is useful in a variety of situations where a sample fluid is to be purified via a diffusion mechanism against an extractor fluid. Moreover, the invention may be used for the removal of components from a sample fluid that vary in size. When blood is the sample fluid, for example, this may include the removal of ‘small’ molecules, ‘middle’ molecules, macromolecules, macromolecular aggregates, and cells, from the blood sample to the extractor fluid.

Claims

1. A device for performing a blood treatment, comprising:

a membraneless processor having a microfluidic channel with a blood inlet, a blood outlet, an extraction fluid inlet, and an extraction fluid outlet,

the blood inlet and the blood outlet having ports for connection to a patient access,

the microfluidic channel being configured such that the blood inlet and outlet and the extraction fluid inlet and outlet are in direct fluid communication without an intervening membrane such that blood and extraction fluid can be placed in direct contact,

the membraneless channel being configured to permit blood and extraction fluid to flow together therein without mixing; and

a membrane-based processor which connects the extraction fluid outlet and the extraction fluid inlet to form a recirculating flow channel,

the extraction fluid outlet being free of any openings or filters effective to block the passage of particles of at least 50,000 Daltons from entering the recirculating flow channel,

the recirculating flow channel being configured such that extraction fluid is able to flow from the extraction fluid outlet to the extraction fluid inlet without passing through a membrane, the membrane-based processor having a membrane positioned along the recirculating flow channel to permit water to flow through the membrane and thereby leave the recirculating flow channel.

2. The device according to claim 1, wherein the microfluidic channel is configured such that it is possible to establish a flow of extraction fluid and blood in which cells in the blood remain substantially separate from cell-free fluid.

3. The device according to claim 1, wherein the microfluidic channel has a ratio of width to height that is more than 10, the width and height being perpendicular to a direction spanning an axis connecting the blood and extraction fluid inlets and the blood and extraction fluid outlets.

4. The device according to claim 1, wherein the microfluidic channel has a height of less than 100 microns, the height being perpendicular to a direction spanning an axis connecting the blood and extraction fluid inlets and the blood and extraction fluid outlets.

5. The device according to claim 1, wherein the extraction fluid inlet opens to the microfluidic channel at two locations that are on either side of the blood inlet, such that a sheath flow that isolates blood from walls of the microfluidic channel can be established therein.

6. A device for performing a blood treatment, comprising:

a microfluidic channel configured such that blood and extraction fluid can be placed in direct contact therein and to permit blood and extraction fluid to flow together therein without mixing; and

a secondary processor which foams a recirculating flow channel between an exit of the microfluidic channel and an entrance of the microfluidic channel, the recirculating flow channel being configured such that extraction fluid from the exit can be recirculated to the entrance without passing through a filter,

the recirculating flow channel including a membrane positioned along a length thereof so as to permit water and uremic toxins in the extraction fluid to leave the recirculating flow channel through the membrane.

7. The device according to claim 6, wherein the recirculating flow channel is configured such that extraction fluid from the exit can be recirculated to the entrance without passing through any openings or filters effective to block the passage of particles of at least 50,000 Daltons.

8. The device according to claim 6, wherein the microfluidic channel is configured such that it is possible to establish a flow of extraction fluid and blood in which cells in the blood remain substantially separate from cell-free fluid.

9. The device according to claim 6, wherein the microfluidic channel has a ratio of width to height that is more than 10, the width and height being perpendicular to a direction spanning an axis connecting the blood and extraction fluid inlets and the blood and extraction fluid outlets.

10. The device according to claim 6, wherein the microfluidic channel has a height of less than 100 microns, the height being perpendicular to a direction spanning an axis connecting the blood and extraction fluid inlets and the blood and extraction fluid outlets.

The device according to claim 6, wherein the microfluidic channel has extraction fluid inlets at two locations at the entrance of the microfluidic channel that are on either side of a blood inlet, such that a sheath flow that isolates the blood from walls of the microfluidic channel can be established therein.

12. The device according to claim 6, wherein the microfluidic channel is configured such that a flow of blood together with an extraction fluid flow therein can be laminar.

13. A device for performing a blood treatment, comprising:

a microfluidic channel configured such that blood and extraction fluid can be placed in direct contact and to permit blood and extraction fluid to flow together therein without mixing; and

a secondary processor configured such that the extraction fluid is able to flow from an extraction fluid outlet of the microfluidic channel on a side of a membrane and back to an extraction fluid inlet of the microfluidic channel without passing through the membrane.

The device according to claim 13, wherein the secondary processor is configured such that extraction fluid can be flowed from the extraction fluid outlet to the extraction fluid inlet without passing through any openings or filters effective to block the passage of particles of at least 50,000 Daltons.

15. The device according to claim 13, wherein the microfluidic channel is configured such that it is possible to establish a flow of extraction fluid and blood in which cells in the blood remain substantially separate from cell-free fluid.

16. The device according to claim 13, wherein the microfluidic channel has a ratio of width to height that is more than 10, the width and height being perpendicular to a direction spanning an axis connecting the blood and extraction fluid inlets and the blood and extraction fluid outlets.

17. The device according to claim 13, wherein the microfluidic channel has a height of less than 100 microns, the height being perpendicular to a direction spanning an axis connecting the blood and extraction fluid inlets and the blood and extraction fluid outlets.

18. The device according to claim 13, wherein the extraction fluid inlet opens to the microfluidic channel at two locations that are on either side of a blood inlet, such that a sheath flow that isolates the blood from walls of the microfluidic channel can be established therein.

19. The device according to claim 13, wherein the microfluidic channel is configured such that a flow of blood together with an extraction fluid flow therein can be laminar.

20. The device according to claim 13, wherein the membrane of the secondary processor is configured to permit water from extraction fluid to flow therethrough.
